几丁聚糖抗自然绝经大鼠卵巢衰老的作用及机制

【摘要】:目的探讨几丁聚糖抗自然绝经大鼠卵巢衰老的作用及机制。方法 30只自然绝经大鼠模型随机分成空白对照组、实验组和雌激素组各10只。实验组给予几丁聚糖1 500 mg/kg灌胃、雌激素组给予雌二醇(E2)1.2 mg/kg灌胃、空白对照组给予生理盐水10 ml/kg灌胃。每天1次,共1个月。采用酶联免疫法测定大鼠血清中孕激素(P)、E2和白细胞介素(IL)-2、IL-6、肿瘤坏死因子(TNF)-α水平,黄嘌呤氧化酶法测定卵巢组织丙二醛(MDA)活力,硫代巴比妥酸法测定卵巢组织超氧化物歧化酶(SOD)的含量。结果 1实验组E2和P高于空白对照组,实验组E2低于雌激素组(P均0.05);2实验组血清IL-2水平明显高于空白对照组,而IL-6、TNF-α水平明显低于空白对照组(P均0.05)。3实验组和雌激素组SOD与空白对照组相比有升高的趋势,但实验组低于雌激素组(P0.05);实验组和雌激素组MDA低于空白对照组(P0.05)。结论几丁聚糖可以通过免疫调节作用和抗氧化作用改善卵巢的内分泌功能,延缓卵巢衰老。
[Abstract]:Objective to investigate the effect and mechanism of chitosan on ovarian aging in natural menopausal rats. Methods Thirty natural menopausal rats were randomly divided into control group (n = 10) and estrogen group (n = 10). The experimental group was given 1 500 mg/kg of chitosan, the estrogen group 1. 2 mg/kg of estradiol, and the control group 10 ml/kg of normal saline. Once a day for 1 month. The levels of serum progesterone E _ 2 and interleukin (IL) -2 IL-6 and tumor necrosis factor TNF- 伪 were measured by enzyme-linked immunosorbent assay (Elisa), and the activity of malondialdehyde (MDA) in ovarian tissue was determined by xanthine oxidase method. The content of superoxide dismutase (SOD) in ovarian tissue was determined by thiobarbituric acid method. Results 1the levels of serum IL-2 in the experimental group were significantly higher than those in the blank control group, and the levels of serum IL-2 in the experimental group were significantly higher than those in the control group. However, the level of IL-6 TNF- 伪 was significantly lower than that of control group (P < 0.05). The SOD of experimental group and estrogen group was significantly higher than that of control group, but the level of MDA in experimental group and estrogen group was lower than that in control group (P 0.05), but the level of MDA in experimental group and estrogen group was lower than that in control group (P 0.05). Conclusion chitosan can improve the endocrine function of ovary and delay ovarian senescence through immunomodulation and antioxidation.
