艾塞那肽对脱氢表雄酮诱导的PCOS大鼠胰岛素抵抗及炎症因子水平的影响

【摘要】:目的探讨艾塞那肽对脱氢表雄酮诱导的多囊卵巢综合征(PCOS)大鼠胰岛素抵抗及炎症因子水平的影响。方法选择SD大鼠30只,皮下注射脱氢表雄酮建立PCOS大鼠模型,随机分为模型组、二甲双胍组和艾塞那肽组,每组10只。另取SD大鼠10只作为正常组。成模后,二甲双胍组、艾塞那肽组分别给予二甲双胍0.265 g/(kg·d)灌胃、艾塞那肽10μg/(kg·d)皮下注射,正常组、模型组给予等量生理盐水灌胃,均连续处理28天。观察各组每周体质量变化。实验结束时行腹腔糖耐量试验,检测空腹血糖(FPG)、餐后2 h血糖(2 h PG)及胰岛素(INS)水平;隔日下腔静脉取血,ELISA法检测血清睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)、性激素结合球蛋白(SHBG)、IL-6、内脂素(visfatin)和色素上皮细胞衍生因子(PEDF)水平。结果与正常组比较,模型组血清T、LH、LH/FSH、FPG、2 h PG、INS、HOMA-IR、IL-6、PEDF水平均显著升高(P均0.05)。与模型组比较,二甲双胍组、艾塞那肽组血清T、LH、LH/FSH、FPG、2 h PG、INS、HOMA-IR、IL-6、PEDF、visfatin水平均显著降低(P均0.05),FSH、SHBG水平均显著升高(P均0.05);二甲双胍组、艾塞那肽组各指标比较差异无统计学意义(P均0.05)。与模型组比较,艾塞那肽组、二甲双胍组体质量均降低(P均0.05),且艾塞那肽组体质量降低的时间更早。结论艾塞那肽在改善PCOS大鼠胰岛素抵抗及降低炎症因子水平方面与二甲双胍相当,但其降低体质量的起效时间较二甲双胍更早。
[Abstract]:Objective to investigate the effects of isenapeptide on insulin resistance and inflammatory factors in rats with dehydroepiandrosterone induced polycystic ovary syndrome (PCOS). Methods Thirty SD rats were randomly divided into three groups: model group, metformin group and isenapeptide group, with 10 rats in each group. Another 10 SD rats were taken as normal group. After the model, metformin group and Isenapeptide group were given metformin 0.265 g/(kg d, Isenapeptide 10 渭 g/(kg d) subcutaneously. The normal group and model group were treated with the same amount of normal saline for 28 days. The changes of weekly body mass in each group were observed. At the end of the experiment, intraperitoneal glucose tolerance test was performed to detect the levels of fasting blood glucose (FPG), 2 h postprandial glucose (PGN) and insulin (ins). The levels of serum testosterone (T), follicle stimulating hormone (FSH), luteinizing hormone (LH), sex hormone binding globulin (SHBG) IL-6 (visfatin) and pigment epithelium-derived factor (PEDFs) were detected by Elisa. Results compared with the normal group, the serum levels of serum TLH / FSHH / FSHFPGN in the model group were significantly higher than those in the control group (P < 0.05). Compared with the model group, the serum levels of TLH / LH / FSHFPGN in metformin group and Eisenapeptide group were significantly lower than those in the metformin group and the isenapeptide group (P < 0.05), but there was no significant difference between the two groups (P = 0.05). Compared with the model group, the body mass of both the Aisenapeptide group and metformin group were all decreased by 0.05%, and the time of the decrease of the body mass in the Isenapeptide group was earlier than that in the control group. Conclusion Isenapeptide is similar to metformin in improving insulin resistance and reducing the level of inflammatory cytokines in PCOS rats, but its effect time of reducing body mass is earlier than that of metformin.
