SDF-1和CXCR4在宫颈癌组织中的表达及临床意义

【摘要】:目的:大量研究发现基质细胞衍生因子-1(SDF-1)和趋化因子受体4(CXCR4)在多种肿瘤的发生发展、浸润转移中发挥着重要作用,,本文旨在探讨SDF-1和CXCR4两种蛋白在宫颈癌中的表达及其临床意义。 方法:应用免疫组织化学染色法,检测80例子宫颈癌,20例正常子宫颈组织中SDF-1和CXCR4蛋白的表达情况;并通过分析比较两者与宫颈癌患者的年龄、病理分级、临床分期、淋巴结转移等方面的关系;研究CXCR4、SDF-1与宫颈癌的发生、发展有无关系,并分析两者之间有无相关性;为宫颈癌的诊断治疗提供一种新的分子靶向治疗的方法。 结果:1.SDF-1蛋白在正常宫颈组织及宫颈癌组织中的表达、SDF-1的表达与宫颈癌的临床病理因素之间的关系 SDF-1蛋白在正常宫颈组织中阳性率为5%(1/20),在宫颈癌组织中表达的阳性率为55%(44/80),二者阳性率的差别有统计学意义(P0.05)。 SDF-1蛋白的表达在宫颈癌临床分期≥Ⅱa期组的阳性率较临床分期Ⅱa期组明显增加(P0.05);有淋巴结转移组的阳性率较无淋巴结转移组明显增加(P0.05);病理分级Ⅰ、Ⅱ、Ⅲ级组的阳性率的差别有统计学意义(P0.05),病理分级Ⅲ级组的阳性率较Ⅰ、Ⅱ级组明显增加(P0.01、P0.01),但病理分级Ⅰ、Ⅱ级组的阳性率无统计学差异(P0.05)。 SDF-1蛋白表达的阳性率与宫颈癌患者年龄大小(P0.05)、原发灶大小(P0.05)、组织类型(P0.05)均无关,无统计学差异。 2. CXCR4蛋白在正常宫颈组织及宫颈癌组织中的表达、CXCR4的表达与宫颈癌的临床病理因素之间的关系 CXCR4蛋白在正常宫颈组织中阳性率为10%(2/20),在宫颈癌组织中表达的阳性率为52.5%(42/80),二者阳性率的差别有统计学意义(P0.05)。 CXCR4蛋白的表达在宫颈癌有淋巴结转移组的阳性率较无淋巴结转移组明显增加(P0.05);临床分期≥Ⅱa期组的阳性率较临床分期Ⅱa期组明显增(P0.05);原发灶大小≥4cm组较原发灶大小4cm组明显增加(P0.05);病理分级Ⅰ、Ⅱ、Ⅲ级组的阳性率差别有统计学意义(P0.05),病理分级Ⅲ级组的阳性率较Ⅰ、Ⅱ级组明显增加(P0.01,P0.01),但病理分级Ⅰ、Ⅱ级组的阳性率无统计学差异(P0.05)。 CXCR4蛋白表达的阳性率与宫颈癌患者年龄大小(P0.05)、组织类型(P0.05)均无关,无统计学差异。 3.宫颈癌组织中SDF-1、CXCR4蛋白表达的相关性 宫颈癌中SDF-1和CXCR4的表达均增加,二者呈正相关性(R=0.473)。 结论:趋化因子CXCR4及其配体SDF-1蛋白的高表达在宫颈癌的恶性增殖、浸润转移有一定意义,它们之间的这种正相关作用可能与宫颈癌的发生发展有一定关系。
[Abstract]:Objective: a large number of studies have found that stromal cell derived factor-1 SDF-1) and chemokine receptor 4 (CXCR4) play an important role in the development, invasion and metastasis of many kinds of tumors. The purpose of this study was to investigate the expression and clinical significance of SDF-1 and CXCR4 proteins in cervical cancer. Methods: immunohistochemical staining method was used to detect the expression of SDF-1 and CXCR4 protein in 20 normal cervical tissues from 80 cases of cervical carcinoma, and to compare the expression of SDF-1 and CXCR4 protein with cervical cancer patients' age, pathological grade and clinical stage. To study the relationship between CXCR4 SDF-1 and cervical cancer, and to analyze the relationship between them, and to provide a new molecular targeted therapy for cervical cancer diagnosis and treatment. Results 1. The expression of SDF-1 protein in normal cervix and cervical carcinoma.
