Beclin-1、Bcl-2、Bax在子宫内膜样腺癌中的表达及意义

【摘要】:【目的】 1.探讨Beclin-1、Bcl-2、Bax三种基因蛋白在子宫内膜样腺癌中的表达及意义。 2.探讨子宫内膜样腺癌中Beclin-1、Bcl-2、Bax三种基因蛋白的表达水平的差异,,三者之间的相关性,以及与临床病理特征之间的关系。 【方法】 运用免疫组化S-P法检测80例子宫内膜样腺癌及40例正常子宫内膜组织制作成的组织芯片中Beclin-1、Bcl-2、Bax三种基因蛋白的表达,并分析三者间的相关性,以及与临床病理特征之间的关系。 【结果】 1、在子宫内膜样腺癌和正常子宫内膜组织中Beclin-1的阳性表达率分别为7.5%和95%;Bcl-2为88.75%和27.5%;Bax为82.5%,20%。Beclin-1在子宫内膜样腺癌以及正常子宫内膜组织中的表达率差异有统计学意义(P0.05)。Bcl-2的表达在子宫内膜样腺癌以及正常子宫内膜组织中的表达率差异有统计学意义(P0.05)。Bax在子宫内膜样腺癌以及正常子宫内膜组织中的表达率差异有统计学意义(P0.05)。 2、本实验子宫内膜样腺癌Bax的表达与Bcl-2的表达无明显相关性(P0.05),Beclin-1与Bcl-2的表达显著负相关(P0.05),Beclin-1与Bax的表达显著负相关(P0.05)。 3、80例子宫内膜样腺癌中Beclin-1的阳性表达率与肿瘤分化程度及TNM分期密切相关(P0.05),与患者年龄、肿瘤大小、浸润深度及淋巴结是否转移均无相关性(P0.05)。Bcl-2的阳性表达率与肿瘤分化程度密切相关(P0.05),组间差异有统计学意义(P0.05),与患者年龄、肿瘤大小、浸润深度、淋巴结是否转移及肿瘤TNM分期均无相关性(P0.05)。Bax的阳性表达率与患者年龄、肿瘤大小、浸润深度、淋巴结是否转移、肿瘤分化程度及TNM分期均无相关性(P0.05)。 【结论】 1、Bax, Bcl-2在子宫内膜样腺癌中高表达,而Beclin-1在子宫内膜样腺癌中低表达,提示三者可能均参与了子宫内膜样腺癌的发生发展。 2、Beclin-1与Bcl-2、Beclin-1与Bax表达呈负相关,提示Bax、Bcl-2可能通过抑制Beclin-1的表达,共同促进子宫内膜样腺癌的发生发展。 3、Beclin-1与Bcl-2的表达与子宫内膜样腺癌的某些临床病理特征之间存在一定相关性。Beclin-1阳性表达与肿瘤分化程度及TNM分期密切相关,可望作为预后评估的参考指标。
[Abstract]:[objective] 1. To investigate the expression and significance of Beclin-1 Bcl-2mBax gene protein in endometrial adenocarcinoma. 2. To investigate the difference of the expression level of three kinds of gene proteins of Beclin-1, Bcl-2 and Bax in endometrial adenocarcinoma, and the correlation between them. Immunohistochemical S-P method was used to detect three kinds of Beclin-1 Bcl-2Bcl-2BX in 80 cases of endometrial adenocarcinoma and 40 cases of normal endometrial tissue. Gene protein expression, The correlation between them and clinicopathological features was analyzed. [results] 1. The positive expression rates of Beclin-1 in endometrioid adenocarcinoma and normal endometrium were 7.5% and 95%, respectively. Bcl-2 was 88.75% and 27.5% respectively. There was significant difference in the expression of Bax in endometrial adenocarcinoma and normal endometrium (P0.05) .Bcl-2 expression in endometrial adenocarcinoma and normal endometrium (P0.05). There was significant difference in the expression rate of Beclin-1 in endometrial adenocarcinoma and normal endometrium (P0.05). The expression rate of Bax in endometrial adenocarcinoma and normal endometrium was significantly different (P0.05). There was no significant correlation between the expression of Bax and Bcl-2 in endometrial adenocarcinoma (P0.05). The expression of Beclin-1 was negatively correlated with the expression of Bax (P0.05). The positive expression rate of Beclin-1 was closely related to tumor differentiation and TNM stage in 80 patients with endometrial adenocarcinoma (P0.05), and correlated with the age of the patients. There was no correlation between tumor size, depth of invasion and lymph node metastasis (P0.05). The positive expression rate of Bcl-2 was closely related to the degree of tumor differentiation (P0.05). The difference between groups was statistically significant (P0.05). There was no correlation between lymph node metastasis and TNM stage (P0.05). The positive expression rate of Bax was correlated with age, tumor size, depth of invasion, lymph node metastasis or not. There was no correlation between tumor differentiation and TNM staging (P0.05). [conclusion] 1Baxand Bcl-2 were highly expressed in endometrial adenocarcinoma, but Beclin-1 was low expression in endometrioid adenocarcinoma. These results suggest that all three may be involved in the development of endometrial adenocarcinoma. 2Beclin-1 and Bcl-2P Beclin-1 are negatively correlated with Bax expression, suggesting that BaxanBcl-2 may inhibit the expression of Beclin-1 by inhibiting the expression of Beclin-1. The expression of Beclin-1 and Bcl-2 was correlated with some clinicopathological features of endometrial adenocarcinoma. The positive expression of Beclin-1 was closely related to the degree of tumor differentiation and TNM stage. It is expected to be a reference index for prognostic evaluation.
