ALDH1作为卵巢癌干细胞标志物的实验研究

【摘要】:目的探讨含生长因子无血清培养基(SFM)培养的卵巢癌细胞株(SKOV3)细胞悬浮球对CD133与乙醛脱氢酶1(ALDH1)表达的影响以及ALDH1是否可作为卵巢癌干细胞(CSC)的标志物。方法用含生长因子的无血清培养基对卵巢癌SKOV3细胞进行培养作为实验组,以含血清培养基(SSM)组作为对照,四甲基偶氮唑蓝比色法(MTT)及平板克隆形成实验、Transwell侵袭实验检测两组细胞的自我增殖、克隆形成及侵袭能力,通过流式细胞技术检测卵巢癌SKOV3细胞和悬浮球中ALDH1、CD133表达情况;体外化疗实验富集ALDH1high细胞初步探索ALDH1在卵巢癌细胞中的作用。结果实验组自我增殖、克隆形成及侵袭能力明显高于对照组(P0.05);实验组ALDH1high、CD133+细胞比例明显高于对照组(P0.05);体外化疗实验表明顺铂连续作用于卵巢癌SKOV3细胞72 h后,实验组ALDH1high、CD133+细胞比例明显增加,ALDH1high细胞比例增加了3.23倍,CD133+细胞比例增加了2.34倍,与对照组相比较差异显著(P0.05)。结论含生长因子的无血清培养基培养卵巢癌SKOV3细胞能够形成肿瘤干细胞球及高表达ALDH1,ALDH1可能是卵巢癌干细胞的标志物之一。
[Abstract]:Objective to investigate the effect of suspension ball on CD133 and ALDH1 expression in ovarian cancer cell line (SKOV3) cultured on serum free medium (SFM) and whether ALDH1 can be used as a marker of ovarian cancer stem cells (CSC). Methods Ovarian cancer SKOV3 cells were cultured in serum-free medium containing growth factor as experimental group and as control group. MTT assay and plate clone formation assay were used to detect the self-proliferation, clone formation and invasion ability of the two groups, and the expression of ALDH1mCD133 in ovarian cancer SKOV3 cells and suspension spheres was detected by flow cytometry. To explore the role of ALDH1 in ovarian cancer cells by chemotherapeutic assay in vitro. Results the self-proliferation, clone formation and invasion ability of the experimental group were significantly higher than those of the control group (P0.05), the percentage of ALDH1-highCD133 cells in the experimental group was significantly higher than that in the control group (P0.05), and the chemotherapeutic experiments in vitro showed that cisplatin continued to act on ovarian cancer SKOV3 cells for 72 hours. The proportion of ALDH1 high CD133 cells in the experimental group was significantly increased by 3.23 times and the proportion of CD133 cells was increased by 2.34 times compared with the control group (P0.05). Conclusion cultured ovarian cancer SKOV3 cells in serum-free medium containing growth factor can form tumor stem cell spheres and overexpression ALDH1-ALDH1 may be one of the markers of ovarian cancer stem cells.
