卵巢子宫内膜异位囊肿患者外周血Th17、Treg比例变化及意义

【摘要】：目的探讨辅助性T细胞17(Th17)、调节性T细胞(Treg)比例及其调控基因在卵巢子宫内膜异位囊肿发生、发展中的作用。方法选择卵巢子宫内膜异位囊肿患者90例作为观察组,因单纯输卵管原因导致的不孕症患者90例作为对照组。采用流式细胞术检测两组外周血Th17、Treg占CD4+T细胞比例,计算Th17/Treg。采用qRT-PCR法检测观察组异位内膜、在位内膜及对照组在位内膜组织孤独核受体γ(ROR-γ)mRNA、叉头状转录因子3(Fox P3)mRNA。采用ELISA法检测两组血清白细胞介素17(IL-17)、IL-22、IL-10、转化生长因子β(TGF-β)。结果观察组外周血Th17占CD4+T细胞比例、Th17/Treg均高于对照组,Treg占CD4+T细胞比例低于对照组(P均0.05)。观察组异位内膜ROR-γmRNA相对表达量低于观察组在位内膜和对照组在位内膜(P均0.05),观察组在位内膜ROR-γmRNA相对表达量低于对照组在位内膜(P0.05);观察组异位内膜Fox P3 mRNA相对表达量高于观察组在位内膜和对照组在位内膜(P均0.05),观察组在位内膜Fox P3 mRNA相对表达量高于对照组在位内膜(P0.05)。观察组血清IL-17、IL-22水平均高于对照组,IL-10、TGF-β水平均低于对照组(P均0.05)。结论卵巢子宫内膜异位囊肿患者外周血Th17/Treg比例失衡;Th17/Treg比例失衡可能通过调节其下游转录因子及细胞因子表达而促进卵巢子宫内膜异位囊肿的发生、发展。
[Abstract]:Objective to investigate the role of helper T cell 17 (Th17), regulatory T cell (Treg) ratio and its regulatory gene in the development of ovarian endometriosis cysts. Methods 90 cases of ovarian endometriosis cyst were selected as observation group and 90 cases of infertility caused by simple fallopian tube as control group. The ratio of Th17,Treg to CD4 T cells in peripheral blood of two groups was detected by flow cytometry, and Th17/Treg. was calculated. Detection of mononuclear receptor 纬 (ROR- 纬) mRNA, forkhead transcription factor 3 (Fox P3) mRNA. in eutopic endometrium, eutopic endometrium and eutopic endometrium by qRT-PCR assay Serum interleukin-17 (IL-17) IL-22 and IL-10, transforming growth factor 尾 (TGF- 尾) were detected by ELISA. Results the ratio of Th17 to CD4 T cells in the observation group was significantly higher than that in the control group (P < 0. 05). The relative expression of ROR- 纬 mRNA in ectopic endometrium in observation group was lower than that in eutopic endometrium of observation group and in eutopic endometrium of control group (P 0.05), and the relative expression of ROR- 纬 mRNA in eutopic endometrium in observation group was lower than that in eutopic endometrium of control group (P0.05); The relative expression of Fox P3 mRNA in eutopic endometrium of observation group was higher than that in eutopic endometrium of control group (P 0.05), and the relative expression of P3 mRNA in eutopic endometrium in observation group was higher than that in eutopic endometrium of control group (P0.05). The level of serum IL-17,IL-22 in the observation group was higher than that in the control group (P < 0. 05). Conclusion the imbalance of Th17/Treg ratio in peripheral blood of patients with ovarian endometriosis and the imbalance of Th17 / Treg ratio may promote the occurrence and development of ovarian endometriosis cysts by regulating the expression of transcription factors and cytokines downstream.
