Livin、Beclin1在卵巢子宫内膜异位症中的表达及意义

【摘要】:目的 通过检测Livin和Beclin1蛋白在卵巢子宫内膜异位症(Ovarian endometriosis,OEMS)患者在位内膜组织、异位内膜组织及对照组正常子宫内膜组织中的表达情况,分析Livin和Beclin1蛋白在各组织中表达的差异及二者之间有无相关性,进一步探讨Livin和Beclin1蛋白在卵巢子宫内膜异位症发生机制中的作用。 研究方法 利用免疫组化染色法检测Livin和Beclin1蛋白在对照组正常子宫内膜和卵巢子宫内膜异位症患者在位内膜、异位内膜组织中的表达,分析上述因子在各组中表达的差异,,应用SPSS13.0软件对所得数据进行统计学处理,采用卡方检验及Fisher’s精确概率法及秩相关分析对其进行统计学分析,以α=0.05作为检验标准,P0.05具有统计学意义。 结果 1. Livin在实验组阳性表达率均高于对照组正常子宫内膜(P0.05),它在对照组正常子宫内膜和实验组在位内膜、异位内膜的阳性表达率分别为18.75%(9/48),60.00%(27/45),86.67%(39/45)。Livin在实验组在位内膜的阳性表达率高于对照组正常子宫内膜(P0.05),在实验组异位内膜的阳性表达率高于在位内膜及对照组正常子宫内膜(P0.05)。Livin在实验组在位内膜、异位内膜增生期与分泌期的表达均无差异(P0.05)。 2. Beclin1在实验组阳性表达率均低于对照组正常子宫内膜(P0.05),它在对照组正常子宫内膜和实验组在位内膜、异位内膜的阳性表达率分别为83.33%(40/48),48.89%(22/45),17.78%(8/45)。Beclin1在在位内膜的阳性表达率低于正常子宫内膜(P0.05),且Beclin1在异位内膜的阳性表达率低于在位内膜和正常子宫内膜(P0.05)。Beclin1在对照组正常子宫内膜组织中分泌期的表达高于增生期(P0.05),而在实验组增生期与分泌期的表达差异无统计学意义(P0.05)。 3.实验组在位内膜和异位内膜组织中Livin基因蛋白表达率Ⅲ-Ⅳ期均高于Ⅰ-Ⅱ期, P0.05,差异有统计学意义;Beclin1基因蛋白表达率Ⅲ-Ⅳ期均低于Ⅰ-Ⅱ期,P0.05,差异有统计学意义。 4. Livin和Beclin1在OEMS中的表达呈负相关。 结论 1. Livin蛋白在在位内膜组、异位内膜组中呈高表达,Beclin1蛋白呈低表达,且与OEMS临床分期关系密切。 2.在OEMS中Livin和Beclin1的表达与月经周期无关。 3. Livin和Beclin1在OEMS中的表达呈负相关,Livin/Beclin1作为一对负向调控蛋白在异位内膜细胞的增殖、凋亡过程中发挥一定作用,共同促进OEMS的发生发展。
[Abstract]:Objective to detect the expression of Livin and Beclin1 protein in eutopic endometrium, ectopic endometrium and normal endometrium of patients with ovarian endometriosis (Ovarian endometriosis,OEMS). To analyze the difference of expression of Livin and Beclin1 in different tissues and the correlation between them, and to explore the role of Livin and Beclin1 in the pathogenesis of ovarian endometriosis. Methods Immunohistochemical staining was used to detect the expression of Livin and Beclin1 protein in eutopic endometrium and ectopic endometrium of normal endometrium and ovarian endometriosis in control group. The differences of the expression of the above factors in each group were analyzed. The data were statistically processed by SPSS13.0 software. The data were statistically analyzed by chi-square test, Fisher's accurate probability method and rank correlation analysis. With 伪 -0.05 as the test standard, P0.05 has statistical significance. The positive expression rate of Livin in the experimental group was higher than that in the normal endometrium of the control group (P0.05), and it was expressed in the normal endometrium of the control group and the eutopic endometrium in the experimental group.
