HIF-1α、CXCR4及VEGF在子宫内膜癌组织中的表达及意义

【摘要】:目的探究缺氧诱导因子(HIF)-1α及其下游基因趋化因子受体(CXCR)4、血管内皮生长因子(VEGF)在子宫内膜癌组织与正常子宫内膜组织中的表达及其意义。方法用逆转录-聚合酶链反应(RT-PCR)及免疫组织化学技术检测94例子宫内膜癌组织和30例正常子宫内膜组织中HIF-1α、CXCR4、VEGF mRNA及蛋白的表达情况,并分析其与子宫内膜癌临床病理特征的关系。结果 HIF-1α、CXCR4、VEGF mRNA及蛋白在子宫内膜癌组织中存在高表达,而在正常子宫内膜组织中几乎无表达。HIF-1α、CXCR4蛋白表达在正常子宫内膜组织中的表达与子宫内膜癌组织比较差异有统计学意义(P0.05)。HIF-1α和CXCR4蛋白的表达水平与子宫内膜癌的脉管浸润、临床分期及患者术后生存时间相关。结论 HIF-1α、CXCR4及VEGF在子宫内膜癌组织中表达水平增高,提示HIF-1α可能通过调控CXCR4及VEGF蛋白的转录,进而促进肿瘤组织新生血管生成而促进子宫内膜癌的发展。
[Abstract]:Objective to investigate the expression and significance of hypoxia inducible factor (HIF) 1 伪 and its downstream chemokine receptor (CXCR) 4 and vascular endothelial growth factor (VEGF) in endometrial carcinoma and normal endometrium. Methods the expression of HIF-1 伪, CXCR4,VEGF mRNA and protein in 94 cases of endometrial carcinoma and 30 cases of normal endometrium were detected by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry. Results HIF-1 伪, CXCR4,VEGF mRNA and protein were highly expressed in endometrial carcinoma. The expression of HIF-1 伪 and CXCR4 protein in normal endometrial tissue was significantly different from that in endometrial carcinoma tissue (P0.05). The expression levels of HIF-1 伪 and CXCR4 protein were correlated with the vascular infiltration of endometrial carcinoma. Clinical stage and postoperative survival time were correlated. Conclusion the expression of HIF-1 伪, CXCR4 and VEGF in endometrial carcinoma is increased, which suggests that HIF-1 伪 may promote the development of endometrial carcinoma by regulating the transcription of CXCR4 and VEGF proteins and thus promoting angiogenesis.
