经γ射线辐照后的脂肪间充质干细胞对大鼠薄型子宫内膜的治疗作用

【摘要】:目的直接进行脂肪间充质干细胞(ADSCs)移植治疗疾病,有可能因为干细胞的无限增殖能力导致治疗的风险性升高,尤其是有致瘤的风险。本实验将γ射线辐照后的ADSCs 移植入小鼠薄型子宫内膜模型,以提高治疗安全性。方法 ADSCs取材于雌性SD大鼠,应用流式细胞技术进行鉴定后,分别以0、5 和10 Gy 的γ射线辐照ADSCs。24 只薄型子宫内膜模型SD大鼠随机分为4 组,在造模6-8 h 后子宫内移植未经辐照的ADSCs(组Ⅰ)、经5 Gy辐照的ADSCs(组Ⅱ)、经10 Gy辐照的ADSCs(组Ⅲ),或仅子宫内注射PBS(组Ⅳ)。测量各组大鼠细胞移植后子宫内膜的厚度变化以观察治疗效果。结果 ADSCs经10 Gy的辐照后完全失去了增殖能力。组Ⅰ和组Ⅱ大鼠在细胞移植治疗后子宫内膜厚度比组Ⅳ有显著增加(P0.01),但是组Ⅲ和组Ⅳ大鼠的子宫内膜厚度无显著差异。结论γ射线破坏了ADSCs的增殖能力,经10 Gy照射后,ADSCs完全失去了增殖能力并失去了治疗能力。经5 Gy照射后的ADSCs部分丧失了增殖能力,但仍有治疗效果,能够改善子宫内膜的厚度。
