干性基因Oct-4和SOX-2在宫颈癌中的表达

【摘要】:目的: 研究干性基因Oct-4和SOX-2在宫颈癌中的表达情况,探讨这二个基因的表达水平能否成为判断宫颈癌愈后的重要指标。 材料与方法: (1)蛋白水平研究:52例宫颈癌标本均取自2008-2012年间吉林大学第一医院妇产科手术治疗患者,病理学检查确诊宫颈癌,所有病例术前均无化疗、放疗和免疫治疗病史。28例正常宫颈组织标本来源于因子宫肌瘤或其他良性疾病行全子宫切除术患者,全部患者无相关癌症病史。通过免疫组织化学SP法检测干性基因Oct-4和SOX-2在宫颈癌和正常宫颈组织中的表达。(2)基因水平研究:选取行宫颈癌根治术患者的新鲜宫颈癌组织12例,,所有病例术前均无化疗、放疗和免疫治疗病史;选取因子宫良性疾病行全子宫切除术患者的新鲜正常宫颈组织6例。通过Q-PCR的方法,检测宫颈癌中Oct-4mRNA和SOX-2mRNA的表达差异。 结果: (1)免疫组织化学染色结果显示:①Oct-4在宫颈癌组织中呈阳性表达,其表达率为46.1%(24/52),显著高于在正常组织中的7.1%(2/28),组间比较差异有统计学意义(p0.005);②SOX-2在宫颈癌组织中呈阳性表达,其表达率为59.6%(31/52),显著高于在正常组织中的10.7%(3/28),组间比较差异有统计学意义(p 0.005);③宫颈癌组织中Oct-4和SOX-2的表达与患者年龄、病理类型和FIGO分期均无关联性;与宫颈癌的分化程度有关,随分化程度的降低Oct-4和SOX-2的表达水平升高,其组间比较差异均有统计学意义,呈负相关;④宫颈癌组织中,Oct-4与SOX-2蛋白的表达具有正相关性关系(p 0.05)。(2)Q-PCR实验结果显示:①SOX-2mRNA在宫颈癌组织中的表达情况(12.67+3.98)高于其在正常宫颈组织的表达情况(1.5+0.548),差异具有统计学意义(t=9.53,p<0.01);②Oct-4mRNA在宫颈癌组织中的表达情况(15.83+3.24)高于其在正常宫颈组织的表达情况(1.333+0.516),差异具有统计学意义(t=15.11,p<0.01);③两者表达未见明显正相关性。 结论: 1在蛋白和mRNA水平上,干性基因Oct-4在宫颈癌组织中表达量均高于正常宫颈组织。 2在蛋白和mRNA水平上,干性基因SOX-2在宫颈癌组织中表达量均高于正常宫颈组织。 3干性基因Oct-4和SOX-2的表达水平可能成为判断宫颈癌愈后的潜在指标。
[Abstract]:Objective: to study the expression of dry gene Oct-4 and SOX-2 in cervical cancer and to explore whether the expression level of these two genes can be an important index to judge the prognosis of cervical cancer. Materials and methods: (1) protein level study: 52 cases of cervical cancer were collected from the patients with gynecology and obstetrics surgery in the first Hospital of Jilin University from 2008 to 2012. The history of radiotherapy and immunotherapy. 28 cases of normal cervical tissues were collected from patients undergoing hysterectomy due to uterine leiomyoma or other benign diseases. The expression of dry genes Oct-4 and SOX-2 in cervical cancer and normal cervix tissues was detected by immunohistochemical SP method. (2) Gene level: 12 cases of fresh cervical cancer tissues with radical cervical cancer resection were selected. All cases had no history of chemotherapy, radiotherapy and immunotherapy before operation. The fresh normal cervical tissues of 6 patients with benign uterine diseases undergoing total hysterectomy were selected. The expression of Oct-4mRNA and SOX-2mRNA in cervical cancer was detected by Q-PCR. Results: (1) Immunohistochemical staining showed that the positive expression rate of 1Oct-4 in cervical carcinoma was 46.1% (24 / 52), which was significantly higher than that in normal tissues (7.1%, 2 / 28). The difference between the two groups was statistically significant (p 0.005). The positive expression rate of 2SOX-2 in cervical cancer tissues was 59.6% (31 / 52), which was significantly higher than that in normal tissues (10.7%, 3 / 28). The difference between the two groups was statistically significant (p 0.005). 3There was no correlation between the expression of Oct-4 and SOX-2 in cervical carcinoma and age, pathological type and FIGO stage. With the decrease of the differentiation degree, the expression of Oct-4 and SOX-2 increased, and the difference between the two groups was statistically significant and negatively correlated with the degree of differentiation. (4) in the tissues of cervical cancer, There was a positive correlation between the expression of Oct-4 and SOX-2 protein (p0.05). (2). The results of Q-PCR assay showed that the expression of 1SOX-2mRNA in cervical cancer tissues (12.67 3.98) was higher than that in normal tissues. Cervical tissue expression (1.5 0.548), The difference was statistically significant (t = 9.53, p < 0.01). The expression of 2Oct-4mRNA in cervical carcinoma (15.83 3.24) was higher than that in normal cervix (1.333 0.516). 3There was no significant positive correlation between the two expressions. Conclusion: 1 at the level of protein and mRNA, the expression of dry gene Oct-4 in cervical carcinoma is higher than that in normal cervix. 2 at the level of protein and mRNA, the expression of dry gene SOX-2 in cervical cancer was higher than that in normal cervix. 3 the expression level of dry gene Oct-4 and SOX-2 may be a potential index to judge the prognosis of cervical cancer.
