敲低KHDRBS3基因抑制人卵巢癌CAOV-3细胞增殖

【摘要】:目的探讨含KH RNA结合结构域信号转导相关分子3(KHDRBS3)基因对体外培养的人卵巢癌CAOV-3细胞增殖的影响。方法取对数生长期细胞,分为未转染组、无关小干涉RNA(siRNA)序列转染组、KHDRBS3-siRNA转染组。采用反转录PCR及Western blot法验证KHDRBS3基因的沉默效果;采用四甲基偶氮唑蓝(MTT)比色法比较不同组别CAOV-3细胞增殖情况;采用流式细胞术检测不同处理组CAOV-3细胞的细胞周期分布。结果 KHDRBS3-siRNA转染后可明显降低CAOV-3细胞KHDRBS3的mRNA和蛋白水平,敲低CAOV-3细胞KHDRBS3水平后,细胞的增殖能力明显降低,G0/G1期细胞比例增加、S期细胞比例降低、G2/M期细胞比例无明显变化。结论敲低KHDRBS3的水平可使CAOV-3细胞发生G0/G1期阻滞,抑制细胞增殖。
[Abstract]:Objective to investigate the effect of signal transduction molecule 3 (KHDRBS3) gene containing KH RNA binding domain on the proliferation of human ovarian cancer CAOV-3 cells in vitro. Methods logarithmic cells were divided into three groups: untransfected group, unrelated small interference RNA (siRNA) sequence transfer group and KHDRBS3-siRNA transfer group. The silencing effect of KHDRBS3 gene was verified by reverse transcription PCR and Western blot, and the proliferation of CAOV-3 cells in different groups was compared by (MTT) colorimetric assay. The cell cycle distribution of CAOV-3 cells in different treatment groups was detected by flow cytometry. Results after KHDRBS3-siRNA transfection, the mRNA and protein levels of KHDRBS3 in CAOV-3 cells were significantly decreased. After knocking down the KHDRBS3 level of CAOV-3 cells, the proliferation ability of CAOV-3 cells was significantly decreased, the proportion of cells in G0/G1 phase was increased, and the proportion of cells in S phase was decreased.
