低氧微环境与宫颈癌耐药的关系

【摘要】:目的通过检测低氧诱导因子-1α(HIF-1α)、多耐药基因1(MDR1)在人宫颈癌组织、He La细胞中的表达及两者间的关系,探讨低氧微环境与宫颈癌耐药的关系。方法应用免疫组化法检测78例中、晚期宫颈癌组织微阵列芯片组织标本中HIF-1α、MDR1的表达;以二氯化钴(Co Cl2)处理He La细胞模拟细胞低氧微环境,设置低氧组、常氧组。细胞免疫荧光法检测HIF-1α、MDR1蛋白在He La细胞中表达。结果 HIF-1α、MDR1在宫颈癌组织中的阳性表达率分别为69.2%、85.6%,且表达呈正相关(r=0.348,P0.05)。低氧组He La细胞HIF-1α、MDR1表达分别为34.76±7.09、57.61±57.61,显著高于常氧组的19.46±3.57、29.57±12.85(P0.05),低氧组HIF-1α与MDR1表达呈正相关(r=0.315,P0.05)。结论低氧微环境与宫颈癌耐药发生有关,可能的机制是HIF-1α促进MDR1的表达从而导致耐药的发生。
[Abstract]:Objective to detect the expression of hypoxia inducible factor-1 伪 (HIF-1 伪) and multi-resistance gene 1 (MDR1) in human cervical cancer, He La cells and their relationship, and to explore the relationship between hypoxia microenvironment and drug resistance in cervical cancer. Methods Immunohistochemical method was used to detect the expression of HIF-1 伪 and MDR1 in 78 cases of advanced cervical cancer tissue microarray tissue, and He La cells were treated with cobalt dichloride (Co Cl2) to simulate the hypoxia microenvironment of the cells, and the hypoxia group and normoxic group were set up. The expression of HIF-1 伪 and MDR1 protein in He La cells was detected by immunofluorescence assay.
