三种修复粘结材料对L929细胞毒性的研究

【摘要】:[目的]通过CCK-8法和流式细胞术的检测,观察了解玻璃离子水门汀、聚羧酸锌水门汀、3M自粘结树脂粘固剂三种粘结材料对小鼠成纤维细胞L929的细胞增殖率和细胞凋亡率的影响及差异,为临床应用提供实验依据。[方法]用CCK-8法检测三种粘结材料浸提3d、7d、14d三个时间段的细胞增殖率(relative growth rate,RGR)和流式细胞术检测三种粘结材料浸提3d、7d两个时间段的细胞凋亡率,观察三种粘结材料对小鼠成纤维细胞L929的影响和差异。[结果]1.玻璃离子水门汀的 RGR 分别为 86.84%(3d)、82.85%(7d)、84.27%(14d),细胞毒性均为1级,表现为轻微细胞毒性。不同浸提时间组之间无明显差异(P0.05)。2.聚羧酸锌水门汀的 RGR 分别为 79.26%(3d)、77.93%(7d)、72.47%(14d),3d和7d细胞毒性为1级,14d细胞毒性为2级,表现为轻微细胞毒性。不同浸提时间组之间无明显差异(P0.05)。3.3M自粘结树脂粘固剂的RGR分别为114.34%(3d)、116.57%(7d)、114.84%(14d),细胞毒性为0级,未表现出细胞毒性。不同浸提时间组之间无明显差异(P0.05)。[结论]1.聚羧酸锌水门汀3d,7d两个时间组的细胞毒性为1级,表现轻微细胞毒性;14d时间组的细胞毒性为2级,表现为轻度细胞毒性;2.玻璃离子水门汀组三个时间组的细胞毒性均为1级,表现出轻微的细胞毒性;3.3M自粘结树脂粘固剂的细胞毒性则为0级,无细胞毒性;4.三种粘结材料均满足口腔材料临床应用的生物相容性的要求。
[Abstract]:[objective] to observe and understand glass ionomer cement by CCK-8 method and flow cytometry. Effects and differences of three kinds of adhesive materials on cell proliferation and apoptosis of mouse fibroblasts L929, which provides experimental basis for clinical application. [methods] the cell proliferation rate and relative growth rateRGRs of three kinds of adhesive materials were measured by CCK-8 and flow cytometry respectively. The apoptotic rates of three kinds of adhesive materials were detected at 3 days and 7 days after extraction. The effects and differences of three kinds of adhesive materials on mouse fibroblasts L 929 were observed. [result] 1. The RGR of vitreous ionomer cement was 86.84 d ~ 82.85 and 84.27 ~ 14 d ~ (-1) respectively. The cytotoxicity of glass ionomer was grade 1, showing slight cytotoxicity. There was no significant difference between different extraction time groups. The RGR of zinc polycarboxylate cement was 79.26 days and 77.93 days, respectively. The cytotoxicity of zinc polycarboxylate was 72.47 days and 14days respectively. The cytotoxicity was grade 1, grade 14, and grade 2, showing slight cytotoxicity. There was no significant difference between different extraction time groups. The RGR of self-adhesive resin binder was 114.343d ~ 116.577d and 114.84d ~ 14d, respectively. The cytotoxicity was 0 grade, no cytotoxicity was found. There was no significant difference between different extraction time groups (P 0.05). [conclusion] 1. The cytotoxicity of zinc polycarboxylate cement for 3 days and 14 days was grade 1 and grade 2, respectively. The cytotoxicity of glass-ionomer cement group was grade 1, and the cytotoxicity of glass-ionomer cement group was grade 0 and no cytotoxicity. All three kinds of adhesive materials meet the requirements of biocompatibility for clinical application of oral materials.
