大蒜油联合顺铂诱导人腺样囊性癌细胞株ACC-M凋亡的研究

【摘要】:目的:探讨大蒜油联合顺铂对人腺样囊性癌细胞株ACC-M细胞增殖、周期及凋亡的影响,为大蒜油、顺铂联合用于临床防治腺样囊性癌提供理论基础和实验依据。 方法:实验分大蒜油组、顺铂组、大蒜油联合顺铂组(1:1),以未用药物处理的ACC-M细胞为空白对照组。人腺样囊性癌ACC-M细胞进行体外培养。MTT法检测2、4、8、16、32μg/mL大蒜油、顺铂、大蒜油联合顺铂作用4、8、12h后人腺样囊性癌细胞株ACC-M体外生长抑制情况;流式细胞仪检测2、8、32μg/mL大蒜油、顺铂、大蒜油联合顺铂作用4、8h后人腺样囊性癌细胞株ACC-M的细胞周期分布和凋亡率情况。 结果:①随着药物浓度增加,各药物组对ACC-M细胞的体外增殖抑制率明显上升(P<0.01);32μg/mL联合组作用12h效果最佳,抑制率为77.16%。大蒜油和顺铂联合用药组抑制癌细胞生长作用明显强于单一用药组(P<0.01)。②大蒜油、顺铂、大蒜油联合顺铂能使ACC-M细胞发生周期分布变化:G0/G1期细胞比例下降,G2/M期细胞比例上升,,S期细胞则无明显变化,呈浓度依赖性(P<0.01)。与单独用药组相比,32μg/mL联合组作用8h作用后G2/M期阻滞更明显(P<0.01)。③随药物浓度增高,大蒜油、顺铂、大蒜油联合顺铂诱导ACC-M细胞凋亡率增高,32μg/mL联合组作用8h凋亡率达7.64%。联合用药组凋亡率明显高于大蒜油和顺铂单独应用组(P<0.01)。 结论:①2、4、8、16、32μg/mL大蒜油、顺铂、大蒜油联合顺铂作用4、8、12h对ACC-M细胞有体外生长抑制作用,存在一定的浓度依耐性;32μg/mL大蒜油联合顺铂作用12h抑制增殖作用明显。②2、8、32μg/mL大蒜油、顺铂、大蒜油联合顺铂作用4、8h能诱导ACC-M细胞株凋亡并阻滞细胞于G2/M期,随药物浓度增加,凋亡率上升,周期分布更明显,32μg/mL大蒜油联合顺铂作用8h效果教佳。③大蒜油联合顺铂应用对ACC-M细胞的作用强于大蒜油、顺铂单独应用。
[Abstract]:Aim: to investigate the effects of garlic oil combined with cisplatin on the proliferation, cell cycle and apoptosis of human adenoid cystic carcinoma cell line ACC-M in order to provide theoretical and experimental basis for the clinical use of garlic oil and cisplatin in the prevention and treatment of adenoid cystic carcinoma. Methods: the experiment was divided into three groups: garlic oil group, cisplatin group, garlic oil combined with cisplatin group (1:1). ACC-M cells were taken as blank control group. Human adenoid cystic carcinoma ACC-M cells were cultured in vitro. The growth inhibition of human adenoid cystic carcinoma cell line ACC-M was detected by MTT assay after 2,4,8,16,32 渭 g / mL garlic oil, cisplatin, garlic oil combined with cisplatin for 4, 8, 12 hours. Flow cytometry was used to detect the cell cycle distribution and apoptosis rate of human adenoid cystic carcinoma cell line ACC-M after 2,8,32 渭 g / mL garlic oil, cisplatin and garlic oil combined with cisplatin for 4 h. Results: 1 with the increase of drug concentration, the inhibition rate of proliferation of ACC-M cells in vitro increased significantly in each drug group, and the inhibition rate was 77.16% in 32 渭 g / mL group for 12 h. The inhibitory effect of garlic oil combined with cisplatin on the growth of cancer cells was significantly stronger than that of single drug group (P < 0.01). 2 garlic oil and cisplatin were found to be more effective in inhibiting the growth of cancer cells (P < 0.01). Garlic oil combined with cisplatin could change the cell cycle distribution of ACC-M cells: the proportion of cells in G0/G1 phase decreased, the proportion of cells in G2 / M phase increased, but the cells in S phase showed no significant change in a concentration-dependent manner (P < 0. 01). The G 2 / M phase arrest was more obvious in 32 渭 g / mL group than that in control group (P < 0 01). 3 the apoptosis rate of ACC-M cells induced by garlic oil, cisplatin and garlic oil combined with cisplatin increased with the increase of drug concentration. The apoptosis rate was 7.64% in 32 渭 g / mL group for 8 h. The apoptosis rate in combination group was significantly higher than that in garlic oil and cisplatin alone group (P < 0.01). Conclusion: 12, 4, 8, 16, 32 渭 g / mL garlic oil, cisplatin, garlic oil combined with cisplatin can inhibit the growth of ACC-M cells in vitro at 4, 8, 12 h, and there is a certain concentration-dependent tolerance. The inhibitory effect of garlic oil (32 渭 g / mL) combined with cisplatin for 12 h was obvious. 22, 8, 32 渭 g / mL garlic oil, cisplatin and garlic oil combined with cisplatin for 4 h and 8 h could induce apoptosis of ACC-M cell line and block cells in G 2 / M phase, with the increase of drug concentration. The apoptosis rate increased and the cycle distribution was more obvious. The effect of combination of 32 渭 g / mL garlic oil and cisplatin for 8 h was better. 3 the effect of garlic oil combined with cisplatin on ACC-M cells was stronger than that of garlic oil, and cisplatin was used alone.
