大蒜油联合5-FU诱导腺样囊性癌细胞株ACC-M凋亡的研究

【摘要】:目的:探讨大蒜油联合5-FU诱导腺样囊性癌细胞株ACC-M凋亡的效应。方法:人涎腺腺样囊性癌细胞株ACC-M细胞培养,分为实验组(5-FU组、大蒜油组、大蒜油+5-FU组)和对照组,采用MTT法观察肿瘤细胞的体外生长活性;采用流式细胞术分析肿瘤细胞周期和凋亡率的变化。结果:MTT实验结果显示不同浓度的5-FU、大蒜油、大蒜油+5-FU对ACC-M细胞均有抑制作用,随着浓度和作用时间的增加而呈上升趋势;流式细胞术分析显示细胞周期发生明显变化,随着浓度增加、作用时间延长G0/G1期的细胞比率上升,S期无明显变化,而G2/M期的细胞下降,凋亡率显示大蒜油联合5-FU诱导ACC-M细胞凋亡明显强于单独组。结论:大蒜油能有效诱导腺样囊性癌细胞株ACC-M凋亡,大蒜油联合5-FU应用对ACC-M细胞的作用明显强于大蒜油,5-FU可单独应用。
[Abstract]:Objective: to investigate the effect of garlic oil combined with 5-FU on apoptosis of adenoid cystic carcinoma cell line ACC-M. Methods: human salivary adenoid cystic carcinoma cell line ACC-M was cultured and divided into experimental group (5-FU group, garlic oil 5-FU group) and control group. The growth activity of tumor cells in vitro was observed by MTT assay. The changes of tumor cell cycle and apoptosis rate were analyzed by flow cytometry. Results: the results of MTT assay showed that garlic oil 5-FU could inhibit ACC-M cells with different concentrations of 5 Fu and garlic oil FU, and increased with the increase of concentration and time. Flow cytometry analysis showed that the cell cycle changed obviously. With the increase of concentration, the cell ratio in G0/G1 phase increased, but there was no significant change in S phase, but decreased in G2 鈮,
