观察GAP-43在骨髓间充质干细胞分化成的类神经细胞中的表达

【摘要】: 目的: 观察β-巯基乙醇及脑源性神经营养因子(brain derived neurotrophic factor,BDNF)诱导下大乳鼠骨髓间充质干细胞向神经样细胞分化后神经生长相关蛋白-43(growth associated protein- 43,GAP-43)的表达变化,以探讨其在骨髓间充质干细胞定向分化过程中的作用。 方法: 体外培养大鼠骨髓间充质干细胞,经流式细胞仪检测细胞表面标志物进行鉴定,分别以β-巯基乙醇及脑源性神经营养因子诱导其向神经元样细胞方向分化,观察诱导后细胞形态学变化,应用免疫细胞化学技术对分化后细胞进行鉴定,分析神经巢蛋白(nestin)、神经元特异性烯醇化酶(NSE)、神经丝蛋白(NF)的表达情况,并观察分化后的GAP-43的表达变化。 结果: 体外培养所得BMSCs经流式细胞仪检测CD90(+)、CD71(+)、CD29(+),CD44(-),经两种方法诱导BMSCs均可分化为神经样细胞,分化后细胞均表达GAP-43、NSE、NF。诱导后1,2,4,6,8小时GAP-43表达阳性率两组均逐渐升高,GAP-43表达强度两组均在诱导后8h内先增强后下降,以诱导后4h表达达到高峰。 结论: 1、骨髓间充质干细胞可以在体外分离及培养。 2、骨髓间充质干细胞可以在体外诱导分化为类神经样细胞。 3、GAP-43在骨髓间充质干细胞向类神经样细胞分化过程中可能起到某种作用甚至是支配作用。
[Abstract]:Aim: to observe the expression of nerve growth related protein 43 (growth associated protein-43 (GAP-43) after differentiation of bone marrow mesenchymal stem cells into neural cells induced by 尾 -mercaptoethanol and brain-derived neurotrophic factor (brain derived neurotrophic). To investigate its role in the differentiation of bone marrow mesenchymal stem cells. Methods: rat bone marrow mesenchymal stem cells were cultured in vitro and identified by flow cytometry. 尾 -mercaptoethanol and brain-derived neurotrophic factor were used to induce the differentiation into neuron-like cells. The morphological changes after induction were observed, the differentiation cells were identified by immunocytochemistry, and the expression of neuron-specific enolase (NSE) and neurofilament protein (NF) of nestin (nestin), were analyzed. The expression of GAP-43 was observed after differentiation. Results: BMSCs cultured in vitro were detected for CD90 () CD71 () and CD29 () CD29 () CD44 (-) by flow cytometry. BMSCs were induced to differentiate into nerve like cells by two methods. The positive rate of GAP-43 expression in both groups increased gradually at 8 hours after induction. The expression intensity of GAP-43 in both groups increased first within 8 hours after induction and then decreased, and reached the peak at 4 hours after induction. Conclusion: 1. Bone marrow mesenchymal stem cells can be isolated and cultured in vitro. 2bone marrow mesenchymal stem cells can be induced to differentiate into neuron-like cells in vitro. Mesenchymal stem cells may play a certain role or even a dominant role in the process of differentiation into neuroblast-like cells.
