百事乐胶囊对慢性应激性抑郁模型小鼠脑内NGF和BDNF的影响

【摘要】:目的: 探讨百事乐胶囊对慢性应激性抑郁模型小鼠行为学及脑中脑源性神经营养因子(BDNF)和神经生长因子(NGF)含量的影响。 方法: 64只雄性昆明种小鼠,随机分为正常组、模型组、百事乐胶囊组和氟西汀组。除正常组以外,其他各组小鼠进行慢性不可预见性应激造模,共造模21d。每天造模前各组小鼠分别给予相应药物灌胃治疗,模型组小鼠以等量蒸馏水灌胃。测定小鼠体质量,通过敞箱实验和悬尾不动实验检测其行为学改变,参考试剂盒采用免疫组织化学法检测小鼠海马BDNF、NGF的含量。 结果: 模型组小鼠发生明显的行为学变化,水平和垂直活动明显减少,悬尾不动时间明显延长;百事乐胶囊组能显著增加小鼠体质量(P0.05),改善小鼠行为学指标(P0.01),百事乐胶囊组能提高海马中BDNF和NGF的含量,与模型组比较差异有统计学意义(P0.05);与氟西汀组相比,百事乐胶囊组各项指标差异无统计学意义(P0.05)。 结论: 百事乐胶囊能显著改善慢性应激性抑郁模型小鼠的行为学变化,其作用可能与提高慢性应激刺激所致的海马BDNF、NGF的含量有关。
[Abstract]:Aim: to investigate the effects of Pepsil capsule on behavior and the contents of brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) in chronic stress depression model mice. Methods: 64 male Kunming mice were randomly divided into normal group, model group, Pepsil capsule group and fluoxetine group. In addition to the normal group, the other groups of mice were induced by chronic unpredictable stress for 21 days. The mice in each group were treated with the corresponding drugs before making the model every day, and the mice in the model group were fed with the same amount of distilled water. The body mass of mice was measured. The behavioral changes were detected by open box test and suspension immobility test. The content of BDNFN NGF in hippocampus of mice was detected by immunohistochemical method with reference kit. Results: in the model group, the behavioral changes were obvious, the horizontal and vertical activities were decreased obviously, and the immobility time of tail suspension was significantly prolonged. The content of BDNF and NGF in hippocampus of the Pepsil capsule group was significantly higher than that of the model group (P0.05), and that of fluoxetine group was higher than that of fluoxetine group (P0.01). There was no significant difference in the indexes of the Pepsil capsule group (P0.05). Conclusion: Pepsil capsule can significantly improve the behavioral changes of chronic stress depression model mice, and its effect may be related to the increase of BDNF NGF content in hippocampus induced by chronic stress stimulation.
