下丘脑室旁核内注射谷氨酸对内脏高敏感性大鼠内脏痛的影响及机制

目的 研究下丘脑室旁核(hypothalamic paraventricular nucleus,PVN)内注射谷氨酸(glutamate,Glu)对慢性内脏高敏感性(chronic visceral hypersensitivity,CVH)大鼠慢性内脏痛的影响及其可能的机制。方法 新生SD大鼠,在出生后第8、10、12天分别给予结直肠扩张法制备CVH大鼠模型。取30只制备成功的CVH模型大鼠,随机分为CVH模型组(CVH组)、CVH+PVN内注射生理盐水组(NS组)、CVH+PVN内注射Glu组3、6、12 μg组(G3组、G6组、G12组),每组6只,另取体质量相匹配的6只SD大鼠作为假手术组(Sham组)。采用痛阈、腹壁撤退反射(abdominal withdrawal reflex,AWR)评分和腹外斜肌肌电图(abdominal external oblique muscle electromyography,EMG)评价大鼠的痛行为。免疫组化染色法检测大鼠脑内精氨酸加压素(arginine vasopressin,AVP)表达水平和结肠组织细胞增殖情况,Tunel法检测结...
