人源重要功能新基因的克

  22周龄人胎肝是胎儿造血的主要场所,此阶段是造血系统迁入和迁出的关键转折点。这一独特性使其极具研究价值。基于对22周龄人胎肝cDNA进行大规模测序的结果,采用生物信息学方法和实验手段(cDNA末端快速扩增法等)获得了13条具有重要功能线索的人源新基因的22条全长cDNA序列。所有这些序列均率先在GenBank注册。关于其中部分基因的主要研究结果如下: 人源Semaphorin 6C和Semaphorin 6D:克隆了Semaphorin基因家族的两个新成员SEMA6C和SEMA6D,它们都属于膜结合型的第六亚家族。这两个基因分别由20个和19个外显子组成,染色体定位分别为1q12-21.1和15q21.1。关于SEMA6C和SEMA6D,分别发现了3种和5种不同剪切形式,它们的表达与组织特异性及发育阶段有关。Northern杂交结果显示,SEMA6C主要在骨骼肌中表达,SEMA6D则在肾、脑和胎盘中高表达,在心肌和骨骼肌中也有一定量的表达。缺失体研究表明,其胞外结构域(Sema结构域和PSI结构域)的完整性是Semaphorin蛋白进行适当的翻译后修饰和亚细胞定位所必须的。SEM...
