成年牛及胎牛β-磷酸三钙煅烧骨骨块的制备、表征和细胞相容性研究

【摘要】:目的:对比研究焦磷酸钠(Na_4P_2O_7.10H_2O,NP)和磷酸二氢铵(NH_4H_2PO_4,MAP)制备的成年牛及胎牛β-TCP煅烧骨骨块的表征及细胞相容性,以期制备出一种理化性能较好且生物安全性可靠的纯相β-TCP煅烧骨骨块。方法:以XRD衍射分析、EDX能谱分析、电镜扫描、Micro-CT扫描及材料力学性能检测研究煅烧骨骨块的表征,以体外细胞毒性实验及细胞与材料的复合实验探究煅烧骨骨块的细胞相容性。结果:NH_4H_2PO_4制备的两种煅烧骨骨块其β-TCP纯度较高,Ca/P比值也均接近于1.47;胎牛煅烧骨骨块的孔隙率、抗压强度及抗弯曲强度均高于成年牛煅烧骨骨块(P0.05);NH_4H_2PO_4制备的两种煅烧骨骨块细胞毒性均为0级,但胎牛煅烧骨骨块上细胞的粘附与增殖速率高于成年牛煅烧骨骨块。结论:NH_4H_2PO_4制备的胎牛β-TCP煅烧骨骨块具有较好的理化性能及细胞相容性,较适用于非承重区骨缺损的修复。
[Abstract]:Objective: to compare sodium pyrophosphate with NaSP _ 4P _ 2O _ (7.10H _ 2O _ (NPN)) and NH _ 4H _ 2PO _ 4 (NH _ 4H _ 2PO _ 4) with ammonium dihydrogen phosphate (NH _ 4H _ 2PO _ 4). The characterization and cytocompatibility of 尾 -TCP calcined bone mass from adult cattle and fetal cattle prepared by MAPA. Objective: to prepare a pure 尾 -TCP calcined bone mass with good physical and chemical properties and reliable biological safety. Methods: edX analysis and scanning electron microscopy (SEM) were used to analyze the bone mass. The characterization of calcined bone mass was studied by Micro-CT scanning and testing of mechanical properties of materials. The cytotoxicity test in vitro and the compound experiment of cell and material were used to study the cytocompatibility of calcined bone mass. Results the purity of 尾 -TCP in the two kinds of calcined bone pieces prepared by 1% NH4H2PO4 was high. The Ca/P ratios were close to 1. 47; The porosity, compressive strength and bending strength of calcinated bone mass of fetal cattle were higher than that of adult calcinated bone mass (P0.05). The cytotoxicity of the two kinds of calcined bone mass prepared by NH_4H_2PO_4 was 0 grade. However, the rate of cell adhesion and proliferation in calcinated bone mass of fetal cattle was higher than that in adult calcinated bone mass. Fetal bovine 尾 -TCP calcined bone mass prepared by NH_4H_2PO_4 has good physicochemical properties and cytocompatibility. It is suitable for the repair of bone defect in non-load-bearing area.
