骨桥蛋白可促进骨关节炎软骨细胞增殖

【摘要】：背景:骨桥蛋白是一种细胞外多功能基质糖蛋白,已被证实参与调控骨关节炎软骨细胞外基质成分的合成分解代谢,但对于骨关节炎软骨细胞本身增殖能力的影响,报道较少。目的:观察骨桥蛋白对人膝骨关节炎软骨细胞增殖能力的影响。方法:所有软骨标本均取自湘雅医院于2012年1至6月因骨关节炎行膝关节置换患者,分离培养骨关节炎软骨细胞,分别予以骨桥蛋白干预、骨桥蛋白sh RNA慢病毒转染、空白对照及Con-shR NA转染等处理,利用MTT及BrdU法检测细胞的增殖能力变化。结果与结论:(1)骨桥蛋白shR NA慢病毒对骨关节炎软骨细胞转染率为80%;(2)与对照组相比,骨桥蛋白干预可明显上调骨关节炎软骨细胞吸光度值(P0.05),而骨桥蛋白sh RNA慢病毒干扰组吸光度值则显著下降(P0.05);(3)骨桥蛋白shR NA转染后,骨桥蛋白的表达水平显著下降(P0.05);(4)结果表明骨桥蛋白可促进骨关节炎软骨细胞增殖,有望成为治疗骨关节炎的新靶点。
[Abstract]:Background: osteopontin is an extracellular multifunctional matrix glycoprotein, which has been proved to be involved in the regulation of the synthesis and catabolism of extracellular matrix components in osteoarthritis chondrocytes, but the effect of osteopontin on the proliferation of osteoarthritis chondrocytes. Objective: to observe the effect of osteopontin on the proliferation of chondrocytes in human knee osteoarthritis. Methods: all cartilage samples were collected from patients undergoing knee arthroplasty in Xiangya Hospital from 2012 to June. Osteoarthritis chondrocytes were isolated and cultured and treated with osteopontin intervention, osteopontin sh RNA lentivirus transfection, blank control and Con-shR na transfection. Results and conclusion the transfection rate of osteopontin shR na lentivirus to osteoarthritis chondrocytes was 802) compared with the control group. Osteopontin intervention significantly upregulated the absorbance of osteoarthritis chondrocytes and decreased the absorbance value of osteopontin sh RNA lentivirus interference group after transfection of osteopontin shR na. The results showed that osteopontin could promote the proliferation of osteoarthritis chondrocytes and might become a new target for the treatment of osteoarthritis.
