氯化镉诱导骨肉瘤细胞凋亡的效应及其分子机制

【摘要】:目的通过观察氯化镉诱导的骨肉瘤细胞凋亡与细胞特异性周期阻滞及caspase活化的影响,阐明氯化镉诱导的细胞凋亡效应及其分子机制。方法应用Annexin V/PI和API等标记,流式细胞仪对人骨肿瘤细胞凋亡及周期特异性的检测,运用RT-PCR的方法检查Caspase-3、Caspase-9活化情况。结果处于静止期的外周血对氯化镉凋亡诱导不敏感,而G_1期氯化镉诱导骨肉瘤细胞出现了明显的细胞凋亡。氯化镉诱导的细胞凋亡主要是在细胞周期的G_1期发生。观察组采用氯化镉凋亡诱导,对照组未做处理,结果显示观察组的Caspase-3、Caspase-9 mRNA的表达值显著高于对照组。结论氯化镉诱导的细胞凋亡与骨肉瘤细胞的细胞周期相关,存在G_1期细胞周期阻滞的周期特异性,且此过程中发生了caspase活化,参与细胞凋亡。
[Abstract]:Objective to investigate the effects of cadmium chloride on apoptosis, cell specific cycle arrest and caspase activation in osteosarcoma cells, and to elucidate the effect of cadmium chloride on apoptosis and its molecular mechanism. Flow cytometry was used to detect the apoptosis and cycle specificity of human bone tumor cells. The activation of Caspase-3 and Caspase-9 was detected by RT-PCR. Results the peripheral blood in stationary phase was not sensitive to the apoptosis induced by cadmium chloride. The apoptosis of osteosarcoma cells was induced by cadmium chloride at the first phase of Gal 1. The apoptosis of osteosarcoma cells induced by cadmium chloride mainly occurred in the first phase of the cell cycle. The observation group was induced by cadmium chloride apoptosis, but the control group was not treated. The results showed that the expression of Caspase-3 and Caspase-9 mRNA in the observation group was significantly higher than that in the control group. Conclusion the apoptosis induced by cadmium chloride is related to the cell cycle of osteosarcoma cells. Participate in apoptosis.
