褪黑素通过下调细胞周期蛋白及其激酶的表达抑制人骨肉瘤MG-63细胞增殖

【摘要】:目的探讨高浓度褪黑素抑制人骨肉瘤细胞系MG-63细胞增殖的潜在机制。方法在体外培养的MG-63细胞中,外源性加入4 mmol/L浓度褪黑素,分别用Western blotting和实时PCR方法检测高浓度褪黑素对细胞周期蛋白(cyclins)及其依赖性激酶(CDK)蛋白和m RNA水平表达的影响。结果 Western blotting和实时PCR检测结果显示,4 mmol/L浓度的褪黑素以时间依赖性方式显著地下调了与细胞周期G_1期进展有关的cyclin D1和CDK4蛋白和m RNA表达水平,与细胞周期G_2/M期进展有关的cyclin B1和CDK1蛋白和m RNA表达水平,但与G_1/S期转变和S期进展有关的cyclin E、CDK2和cyclin A蛋白和m RNA表达水平没有变化。结论褪黑素以时间依赖性方式,通过诱导细胞周期停滞抑制骨肉瘤MG-63细胞增殖,这种抑制效应与cyclin D1和CDK4及cyclin B1和CDK1的表达下调有关。
[Abstract]:Objective to investigate the potential mechanism of high concentration of melatonin in inhibiting proliferation of human osteosarcoma cell line MG-63. Methods in vitro cultured MG-63 cells were treated with 4 mmol/L melatonin. Western blotting and real time PCR were used to detect the effects of melatonin on the expression of cyclin cyclins and their dependent kinases, and the expression of m RNA. Results Western blotting and real time PCR assays showed that melatonin 4 mmol/L concentration was melatonin. The expression levels of cyclin D1 and CDK4 proteins and m RNA related to the progression of cell cycle G1 were significantly down-regulated in a time-dependent manner. The expression levels of cyclin B1 and CDK1 protein and m RNA related to the progression of G2 / M phase in cell cycle were not changed, but the expression levels of cyclin ECDK2 and cyclin A protein and m RNA related to G1- S phase transition and S phase progression were not changed. Conclusion melatonin is time-dependent. The proliferation of osteosarcoma MG-63 cells was inhibited by cell cycle arrest, which was related to the down-regulation of the expression of cyclin D1 and CDK4 and cyclin B1 and CDK1.
