AQP3、AQP8在大鼠结肠大部切除术后的代偿情况及药物对其影响的研究

【摘要】:目的:通过观测大鼠行结肠大部切除术后剩余结肠中水通道蛋白(Aquaporin AQP)3、8的表达情况及药物对其的影响,研究术后剩余结肠是否有代偿性增加水通道蛋白3、8的表达以增强结肠水分吸收的作用,并探讨痛泻要方、盐酸洛哌丁胺的止泻作用是否与促进水通道蛋白3、8的表达有关,从而为临床上结肠切除术后发生腹泻提供诊疗依据和方法。方法:选取成年雄性SD大鼠36只,按照随机分组原则分为空白组(A组)、模型组(B组)、痛泻要方干预实验组(C组)、盐酸洛哌丁胺干预实验组(D组),每组9只。其中,A组大鼠单笼常规饲养;B-D组在全麻下行结肠大部切除端端吻合术,术后均单笼常规饲养,予抗感染治疗3天;C组、D组于术后第一天起分别予痛泻要方、盐酸洛哌丁胺灌胃1周。每组分别于术后第1、2、4周随机取3只大鼠结肠吻合口处肠道组织,A组取临近手术部位肠道组织。再按照免疫组化学和实时荧光定量聚合酶链式反应(Real-Time RT-PCR)的方法及要求对结肠组织中AQP3、8的表达进行检测,最后用统计学的方法对检测结果进行比较。结果:1.与A组相比,B组、C组、D组大鼠剩余结肠组织AQP3、8表达均上调,差异有统计学意义(p0.01)。2.与B组相比,C组、D组大鼠剩余结肠组织AQP3、8表达含量均上调,差异有统计学意义(p0.05)。3.与D组相比,C组大鼠剩余结肠组织AQP3、8表达含量均上调,差异有统计学意义(p0.05)。结论:大鼠结肠大部切除术后剩余结肠AQP3、8的表达上调,且其活跃程度与术后时间有相关性;痛泻要方、盐酸洛哌丁胺对结肠大部切除术后剩余结肠AQP3、8的表达有明显的促进作用,且痛泻要方的促进作用较盐酸洛哌丁胺更明显。
[Abstract]:Objective: to observe the expression of aquaporin aquaporin aquaporin (Aquaporin) in the residual colon of rats after subtotal colectomy and the effect of drugs on it. To study whether there is a compensatory increase in the expression of aquaporin 3O8 in the residual colon after operation to enhance the water absorption of colon, and to explore whether the antidiarrhea effect of Lupibutylamine Hydrochloride is related to the promotion of the expression of aquaporin 3O8. Methods: 36 adult male Sprague-Dawley rats were selected. According to the principle of random grouping, they were divided into blank group A group, model group B group, Tongxie Yaofang intervention group C group, and loperamine hydrochloride intervention group D group, 9 rats in each group. Descending subtotal resection of colon end-to-end anastomosis, All the patients were fed with single cage after operation and treated with anti-infective therapy for 3 days. Group D was given Tongxie decoction from the first day after operation. Each group was randomly selected from intestinal tissue of colon anastomosis site of 3 rats in 4 weeks after operation. The intestinal tissue of group A was taken from adjacent site of operation. Then according to immunohistochemistry and real-time fluorescence quantitative polymerase, the intestinal tissue of each group was obtained by intragastric administration of lopipramine hydrochloride for 1 week. The method of chain reaction Real-Time RT-PCR was used to detect the expression of AQP3p8 in colonic tissue. Finally, the results were compared with those in group A. results compared with group A, the expression of AQP3p8 in the residual colon of group B and C was up-regulated, and the expression of AQP3p8 in group D was up-regulated. Compared with group B, the expression of AQP3O8 in group D and group C were all up-regulated, and the difference was statistically significant (P < 0.05). Compared with group D, the expression of AQP3O8 in residual colon of group C was up-regulated, and that of group C was significantly higher than that of group C (P < 0.05). Conclusion: after subtotal colectomy, the expression of AQP3O8 in the residual colon of rats is up-regulated, and its activity is correlated with the postoperative time. Lupibutylamine hydrochloride promoted the expression of AQP3O8 in residual colon after subtotal colectomy, and the promoting effect of Tongxieyao recipe was more obvious than that of Lupibutylamine hydrochloride.
