硫酸氨基葡萄糖对口服透明质酸缓解关节炎症的增效作用

【摘要】:目的:关节炎是现代社会中常见的一种慢性疾病,在不同年龄层次的人群中,都有相当高的比例,并且逐年都在增加。这种慢性疾病已经给人们的生活带来了许多负面的影响。很多关节炎患者选择关节内注射透明质酸的方式,治疗疾病,但因为关节注射会带来巨大的疼痛感,导致一大部分患者无法忍受,从而放弃治疗。透明质酸和硫酸氨基葡萄糖作为天然产物,存在于人体关节组织内,是非常重要的组成成分,并且作为功能性食品,其食用价值已经得到国内外的广泛认可。本研究就结合国内外的研究报道,从三方面去探讨联合口服透明质酸和硫酸氨基葡萄糖对关节炎症的缓解作用:(1)在透明质酸中加入硫酸氨基葡萄糖,对其基础理化性质进行测定。(2)研究硫酸氨基葡萄糖对口服透明质酸缓解关节炎症的增效作用。(3)通过3种浓度配比的HA复合液对比,探究HA+GS(2:1)对关节炎症缓解作用的有效性。方法:针对上述研究,设计一下试验方法:(1)以透明质酸纯物质为标准,测定HA复合液中pH值、水分含量、粘度与D-葡萄糖醛酸的含量,观察加入硫酸氨基葡萄糖后,HA的基本理化性质是否受到影响。(2)将50只8周龄ICR雄性小鼠随机分成5组,正常组、模型组、HA组、GS组和HA+GS组。除正常组以外,其余4组小鼠左后足注射弗氏完全佐剂(freund’s complete adjuvant,FCA)建立佐剂性关节炎模型,然后进行4周的HA和GS口服给药,并分别对小鼠体质量、足跖厚度、宽度、肿胀度及脚踝肿胀度进行测定,同时采用酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)检测小鼠血清中白细胞介素-1(interleukin-1,IL-1)及肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的含量。(3)将50只8周龄ICR雄性小鼠按照体重平均分为5组:正常、模型、HA+GS(2:1)、HA+GS(1:2)、HA+GS(1:1)。除正常组外,对其余4组小鼠进行FCA造模,继续饲养10天后,对小鼠进行尾部取血,然后连续口服给药30天。在此期间,对小鼠足跖厚度和宽度进行测量,并在口服结束后,解刨取血,采用酶联免疫吸附法检测给药前和给药后的白介素1(IL-1)的含量。结果:(1)HA纯物质与HA复合物的各项指标测定结果为:p H值6.30、6.09;水分含量5.47%、5.84%;粘度209.59、209.91;D-葡萄糖醛酸的含量49.73%、48.23%。(2)口服HA和GS都能够减轻小鼠足跖与脚踝的肿胀现象,而同时口服HA和GS能起到更好的消肿效果,血清中的IL-1及TNF-α的含量也显著降低。(3)通过对小鼠足跖肿胀度差值以及足跖肿胀度变化值的分析,发现口服3种浓度配比的HA混合液后,足跖肿胀度差值,足跖肿胀度变化值以及IL-1的含量都有明显的降低,且这3组与模型组之间存在显著性差异(p0.05);其中,HA+GS(2:1)和HA+GS(1:2)组的降低程度相对于HA+GS(1:1)组更为显著,它们之间存在显著性差异(p0.05)。结论:(1)透明质酸中加入硫酸氨基葡萄糖后,对HA纯物质的性质并未造成显著的改变,依然符合HA相关国家规定。(2)给予小鼠口服HA+GS,比单独服用HA、GS能更有效、更快地缓解小鼠的足跖肿胀症状,同时能够降低血清中IL-1及TNF-α含量,进而对小鼠佐剂性关节炎起到更好的治愈效果。(3)口服3种不同浓度配比的HA混合液能够减轻佐剂性小鼠足跖肿胀度,并降低血液中IL-1的含量,对关节炎症起到了一定的缓解效果,但这3种浓度配比的透明质酸混合液组效果存在一定差异但并不显著,其中HA+GS(2:1)对关节炎症的缓解效果明显好于HA+GS(1:1),但与HA+GS(1:2)之间的效果差异并不显著。
[Abstract]:Objective: arthritis is a common chronic disease in modern society, in different age groups, have a very high proportion, and are on the increase year by year. Brought many negative effects of this chronic disease has brought to people's lives. Many arthritis patients choose the way of intraarticular injection of hyaluronic acid the treatment of the disease, but because the joint injection will bring huge pain, resulting in a majority of patients can not endure, to give up treatment. Hyaluronic acid and glucosamine sulfate as a natural product, exist in the human body joint organization, is a very important component, and as functional foods, edible value has been widely recognized at home and abroad. The research on the combination of research reports at home and abroad, to explore the combined oral hyaluronic acid and glucosamine sulfate on relieving inflammation in the joints from three aspects: (1) in Adding glucosamine sulfate and hyaluronic acid, on the basis of physico-chemical properties were measured. (2) study of glucosamine sulfate on oral hyaluronic acid to alleviate synergism of joint inflammation. (3) through the comparison of 3 kinds of HA composite liquid ratio HA+GS (2:1), to explore the effectiveness of mitigation effect on arthritis disease. Methods: according to the above research, test method design: (1) with hyaluronic acid as the standard of pure substances, determination of HA composite liquid pH value, moisture content, viscosity and D- content of glucuronic acid, treated with glucosamine sulfate, basic physicochemical properties of HA are affected (2). 50 8 week old male ICR mice were randomly divided into 5 groups, normal group, model group, HA group, GS group and HA+GS group.
