六磷酸肌醇对人骨肉瘤细胞株MG-63和原代培养成骨细胞增殖的影响

【摘要】:目的研究六磷酸肌醇(inositol hexaphosphate,IP6)对人骨肉瘤细胞株MG-63和原代培养成骨细胞增殖的影响,并探求其抗癌效果的最佳作用浓度。方法原代培养人成骨细胞并鉴定,复苏并常规培养人骨肉瘤细胞株MG-63,MTT法检测不同浓度IP6干预培养时两种细胞的增殖情况,确定IP6的最佳抑癌浓度,并进一步用流式细胞仪检测MG-63细胞周期及凋亡情况。结果当IP6浓度≥1mmol/L时开始抑制MG-63细胞增殖,在4mmol/L时达到最大,并呈时间-剂量依赖性;当IP6浓度为0.5、1mmol/L时,对成骨细胞增殖抑制不明显,2mmol/L时轻度抑制,4mmol/L时导致成骨细胞凋亡。结论 IP6能够抑制人骨肉瘤细胞MG-63的增殖并导致其凋亡,其抗癌的最佳作用浓度为2mmol/L。
