右美托咪定给药后引起大鼠心脏MicroRNA差异表达

【摘要】:目的:右美托咪定的应用可能在心脏外科手术中具有有利影响。MicroRNAs是一种有基因表达调控功能的非编码RNA。本研究旨在研究MicroRNAs在右美托咪定心脏保护中的作用。方法:本研究入选了六只大鼠并分成了两组:右美托咪定组和对照组,右美托咪定组大鼠腹腔注射右美托咪定(100ug/Kg),对照组大鼠腹腔注射同等剂量的生理盐水。给射后30分钟,麻醉下取出大鼠标记后储存在液氮中。所有样本的microRNAs表达量通过microRNA基因芯片检测。结果:两组大鼠心脏microRNA表达量存在差异,其中5 个microRNA(mo-miRNA-X2,rno-miRNA-X3,rno-miRNA-X4,rno-miRNA-X5 and rno-miRNA-X6)的表达量上调的,1个microRNA的表达量是下调的。对照组和右美托咪定组的大鼠的microRNAs表达量通过T检验进行对比,P小于0.05(P0.05),这表明上述结果具有统计学意义。结论:右美托咪定可引起心脏microRNA产生差异表达,这些差异表达的microRNA可能与心脏保护有关。
[Abstract]:Objective: the application of dexmetomidine may have a beneficial effect in cardiac surgery. MicroRNAs are a kind of non-coding RNAs with the function of gene expression regulation. The purpose of this study was to investigate the role of MicroRNAs in the cardiac protection of dexmetomidine. Methods: six rats were divided into two groups: right metoimidine group and control group, right metoimidine group rats were injected intraperitoneally with dexmetoimidine 100ugr / KgG, and control rats were intraperitoneally injected with the same dose of normal saline. 30 minutes after injection, the rats were taken out under anesthesia and stored in liquid nitrogen. The expression of microRNAs in all samples was detected by microRNA gene chip. Results: there was significant difference in the expression of microRNA between the two groups. The expression of 5 microRNAs mo-miRNA-X2nrno-miRNA-X3rno-miRNA-X4 rno-miRNA-X5 and rno-miRNA-X6 was up-regulated, and one microRNA was down-regulated. The expression of microRNAs in the control group and dexmetidine group was compared by T test (P < 0.05), which indicated that the above results were statistically significant. Conclusion: dexmetomidine can induce differential expression of microRNA in the heart, and these differential expressions of microRNA may be related to cardiac protection.
