皮肤扩张早期miRNA和mRNA的变化及关键miRNA筛选

【摘要】:目的建立皮肤扩张模型,探究皮肤扩张早期中miRNA和mRNA表达谱的变化情况。方法取6-8周SD大鼠建立扩张模型,在扩张早期取下扩张皮肤,并利用未扩张皮肤作为对照,采用Affymetrix microRNA 3.0平台和Affymetrix GeneChip Rat Gene 1.0 ST检测扩张早期miRNA和mRNA的表达变化;利用统计学方法初步筛选出表达发生变化的miRNA和mRNA,并根据差异表达的倍数筛选出表达差异较大的关键miRNA。采用TargetScan、miRDB和IPA数据库对关键miRNA进行靶点预测;利用miRNA与靶点mRNA数量变化呈负相关的关系初步明确扩张早期miRNA可能调控的mRNA靶点,建立miRNA-靶mRNA的一一对应关系。对靶点mRNA进行功能分析和通路分析,明确扩张早期关键miRNA-靶mRNA-通路的对应关系。结果筛选出了11个满足统计学差异和表达倍数变化的miRNA,明确了miRNA-靶mRNA之间的相互作用关系;这些miRNA调控的靶点参与TGF-βSignaling、Apoptosis Signaling和PDGF Signaling等通路。结论本研究筛选出了11个皮肤扩张早期的关键miRNA,调控扩张过程中的关键通路,这些miRNA可作为调控皮肤早期扩张的靶点。
[Abstract]:Objective to establish a skin dilatation model and investigate the changes of miRNA and mRNA expression profiles in the early stage of skin dilatation. Methods Sprague-Dawley rats (6-8 weeks old) were used to establish dilatation model. The expression of miRNA and mRNA were detected by Affymetrix microRNA 3.0 platform and Affymetrix GeneChip Rat Gene 1.0ST in the early stage of dilatation. The expression of miRNA and mRNAs were preliminarily screened by statistical method, and the key miRNAs were screened according to the multiple of differentially expressed miRNAs. The target of miRNA was predicted by TargetScanmiRDB and IPA database, and the relationship between miRNA and the quantity of mRNA was negatively correlated, and the mRNA target which might be regulated by miRNA in the early period of expansion was identified, and the one-to-one correspondence of miRNA-target mRNA was established. The functional analysis and pathway analysis of target mRNA were carried out to clarify the relationship between the key miRNA-target mRNA-pathway in the early stage of expansion. Results Eleven miRNAs satisfying the statistical difference and the change of expression multiple were screened, and the interaction between miRNA-target mRNA was clarified, and these miRNA regulatory targets were involved in the pathway of TGF- 尾 signaling Signaling and PDGF Signaling. Conclusion in this study, 11 key miRNAs in the early stage of skin dilatation were screened and the key pathways in the process of skin expansion were regulated. These miRNA can be used as targets for early skin dilatation.
