兔前交叉韧带损伤后残端血运变化的研究

【摘要】:目的:探讨兔前交叉韧带(anterior cruciate ligament,ACL)损伤后残端血运的变化情况,为ACL保残重建提供进一步的依据。方法:新西兰大白兔共30只,雌雄不限,随机分成5个组,每个组6只,其中1组作为空白对照组,其余各组分别将其右后肢ACL自股骨端切断,建立ACL损伤动物模型。分别于损伤后2周、4周、8周、12周时处死实验动物,采集ACL残端标本。将标本进行HE染色组织观察、免疫组织化学染色计算微血管密度。结果:与空白对照组相比,损伤后第2周、4周残端微血管数量均升高(P0.05),且4周时高于其他组(P0.05)。损伤后8周微血管数量开始下降,至12周时降到最低,低于空白对照组(P0.05)。结论:1.ACL残端血运随时间变化具有明显差异性,呈现先增高后降低的趋势。2.根据残端血运变化情况提示,越早进行保留ACL残端重建,对移植物再血管化就越有利。
[Abstract]:Objective: to investigate the changes of stump blood flow after anterior cruciate ligament (ACL) injury in rabbits. Methods: thirty New Zealand white rabbits were randomly divided into 5 groups with 6 rabbits in each group. The first group was used as the blank control group. The other groups cut the right hind limb ACL from the femur to establish the ACL injury animal model. The experimental animals were killed at 2 weeks, 4 weeks, 8 weeks and 12 weeks after injury, and ACL stump specimens were collected. The specimens were observed by HE staining and microvessel density was calculated by immunohistochemical staining. Results: compared with the control group, the number of microvessels in the stump increased at the 2nd week after injury (P0.05), and was higher than that in the other groups at the 4th week (P0.05). The number of microvessels began to decrease at 8 weeks after injury and reached the lowest at 12 weeks, which was lower than that in the blank control group (P0.05). Conclusion: 1. ACL stump blood flow has obvious difference with time, showing a trend of increase first and then decrease. 2. According to the changes of stump blood flow, the earlier the preservation of ACL stump, the more favorable for graft revascularization.
