成软骨预分化处理对脂肪干细胞在退变椎间盘中再生修复作用的影响

【摘要】:目的近年来,干细胞移植逐步成为椎间盘退变性疾病治疗的研究热门。退变椎间盘中恶劣的微环境对移植干细胞的生长、增殖、分化都有影响。近期有研究表明,"预分化"处理可以增强移植细胞对微环境的适应能力。本研究旨在探究预分化处理对干细胞在椎间盘微环境中生物学行为的影响。方法将大鼠脂肪干细胞(ADSC,adipose-derived stem cell)分为5组,予Ⅱ型胶原(typeⅡcollagen)包埋处理后,分别进行成软骨分化处理0天(对照组),3,7,10,14天(实验组);再将各组细胞移植于模拟退变椎间盘的微环境(酸性,低糖,高渗透压)培养7天;用细胞计数试剂盒(CCK-8)检测各组细胞的增殖情况,用聚合酶链式反应(RT-PCR)、蛋白印记及免疫荧光法检测干细胞的细胞外基质合成情况。结果一周的分化过程中,预分化7天的ADSC增殖能力相对最强。功能基因合成方面,D7组和D10组Ⅱ型胶原mRNA的表达量均较对照组增高;D7组的多聚蛋白聚糖mRNA表达量较对照组高。结论通过本次研究我们得出结论,成软骨细胞预分化处理7至10天的可能促进脂肪干细胞在椎间盘恶劣环境中的增殖能力和细胞外基质合成能力,有利于干细胞对退变椎间盘的再生。
