音猬因子联合甲基强的松龙治疗大鼠脊髓损伤的实验研究

【摘要】:目的:本课题旨在探讨音猬因子(Shh)联合甲基强的松龙(MP)的治疗方法,治疗大鼠脊髓损伤的实验研究。方法:本次研究选取40只雌性SD大鼠,体重均为200-250g。采用改良Allen打击法制备大鼠急性脊髓损伤模型,蛛网膜下腔置管加尾静脉注射两者给药方式。将实验动物随机分为4组,每组10只:空白对照A组采用造模后24h内尾静脉注入生理盐水+蛛网膜下腔注入生理盐水连续30天;单一治疗B组采用造模后24h内尾静脉注入MP+蛛网膜下腔注入生理盐水连续30天;单一治疗C组采用造模后24h内尾静脉注入生理盐水+蛛网膜下腔注入shh连续30天;联合治疗D组采用造模后24h内尾静脉注入MP+蛛网膜下腔注入shh连续30天。在造模成功后每周均对各组实验动物行BBB评分,直至造模成功后第五周以评价神经运动功能恢复情况;在BBB评分结束后处死各组实验动物,取材行HE染色比较两组大鼠脊髓损伤后病理变化。结果:BBB评分:联合治疗组BBB评分明显高于其他实验三组,且差异具有统计学意义(P0.05);MP组BBB评分高于空白对照组以及Shh组,差异具有统计学意义(P0.05);Shh组较空白对照组BBB评分改变不明显,差异不具有统计学意义(P0.05)。HE染色显示联合治疗组较其他实验三组脊髓损伤后空洞面积明显缩小,且差异具有统计学意义(P0.05);MP组脊髓损伤后空洞面积小于空白对照组以及Shh组,差异具有统计学意义(P0.05);Shh组较空白对照组脊髓损伤后空洞面积缩小不明显,差异不具有统计学意义(P0.05)。结论:1.音猬因子单独使用对大鼠脊髓损伤后功能的恢复作用不明显,但甲基强的松龙单独使用可促进大鼠脊髓损伤后功能的恢复;2.音猬因子联合甲基强的松龙治疗可以明显改善脊髓损伤后的功能恢复。
[Abstract]:Aim: to investigate the therapeutic effect of (Shh) combined with methylprednisolone (MP) on spinal cord injury (sci) in rats. Methods: 40 female Sprague-Dawley rats (200-250 g) were selected in this study. The model of acute spinal cord injury in rats was established by modified Allen method. The two drugs were administered by subarachnoid tube and caudal vein injection. The experimental animals were randomly divided into 4 groups with 10 rats in each group: control group A was injected with saline subarachnoid intravenously for 30 days within 24 hours after modeling. In group B, saline was injected into subarachnoid cavity of MP within 24 hours after modeling, and shh was injected into subarachnoid space of tail vein in group C for 30 days. Group D was treated with injection of shh into subarachnoid cavity of MP within 24 hours after modeling. Every week after the success of the model, the experimental animals were evaluated with BBB score until the fifth week after the model was made, and the experimental animals were killed after the BBB score was finished. The pathological changes after spinal cord injury in two groups were compared by HE staining. Results: the BBB score of the combined treatment group was significantly higher than that of the other three experimental groups, and the difference was statistically significant (P0.05) the BBB score of MP group was higher than that of the blank control group and Shh group. The difference was statistically significant (P0.05) the BBB score in the Shh group was not significantly different from that in the blank control group (P0.05). The HE staining showed that the cavity area in the combined treatment group was significantly smaller than that in the other three experimental groups. The difference was statistically significant (P0.05). The area of cavities after spinal cord injury in MP group was smaller than that in control group and Shh group. The difference was statistically significant (P0.05). Conclusion 1. The effect of Hedgehog on the recovery of function after spinal cord injury in rats was not obvious, but methylprednisolone alone could promote the recovery of function after spinal cord injury in rats. Hedgehog factor combined with methylprednisolone can significantly improve the functional recovery after spinal cord injury.
