ADAMTS-7、ADAMTS-12在退变腰椎间盘组织中的表达及临床意义

【摘要】:【目的】检测ADAMTS-7、ADAMTS-12在人体腰椎间盘髓核组织中的表达情况,探讨ADAMTS-7、ADAMTS-12在人体腰椎间盘髓核组织退变中的作用,为研究人体腰椎间盘退变的机制提供一种新的研究方向。【方法】实验组40例,为手术切除腰椎间盘突出症患者退变的椎间盘组织,其中后外层纤维环破裂22例,后外层纤维环未破裂18例;对照组10例,为腰椎骨折患者正常的腰椎间盘组织;采用半定量RT-PCR和免疫组织化学技术分别从m RNA和蛋白质水平检测标本中ADAMTS-7、ADAMTS-12的表达情况,进行统计学分析。【结果】ADAMTS-7、ADAMTS-12蛋白质表达含量在后外层纤维环破裂组和后外层纤维环未破裂组均明显高于正常组,后外层纤维环破裂组又明显高于后外层纤维环未破裂组(P0.01);ADAMTS-7、ADAMTS-12 m RNA表达量在后外层纤维环破裂组和后外层纤维环未破裂组均明显高于正常组,后外层纤维环破裂组又明显高于后外层纤维环未破裂组(P0.01)。【结论】ADAMTS-7、ADAMTS-12均有可能参与人体腰椎间盘组织退变的病理过程,有一定的临床研究意义。且随着人体腰椎间盘组织退变程度的增加,ADAMTS-7、ADAMTS-12的表达逐渐升高。
[Abstract]:[objective] to detect the expression of ADAMTS-7 and ADAMTS-12 in the nucleus pulposus of human lumbar disc, and to explore the role of ADAMTS-7 and ADAMTS-12 in the degeneration of the nucleus pulposus of human lumbar intervertebral disc. To provide a new research direction for studying the mechanism of degeneration of human lumbar intervertebral disc. [methods] Forty patients with lumbar disc herniation underwent surgical excision of degenerated intervertebral disc tissue, 22 of them had posterior outer fibrous ring rupture. There were 18 cases of unruptured posterior outer fibrous ring and 10 cases of control group, which were normal lumbar intervertebral disc tissue. The expression of ADAMTS-7 and ADAMTS-12 was detected by semi-quantitative RT-PCR and immunohistochemistry from m RNA and protein levels, respectively. [results] the protein expression of ADAMTS-7 and ADAMTS-12 was significantly higher in the group of the rupture of the posterior outer fiber ring and the group of the non-rupture of the posterior outer fiber ring than that of the normal group. The expression of ADAMTS-7 and ADAMTS-12 m RNA in the ruptured posterior outer fibrous ring group was significantly higher than that in the unruptured posterior outer fibrous ring group (P0.01), and the expression of ADAMTS-12 m RNA in the posterior outer fibrous ring rupture group and the posterior outer fibrous ring unruptured group was significantly higher than that in the normal group. ADAMTS-7 and ADAMTS-12 may be involved in the pathological process of degeneration of human lumbar intervertebral disc, which has some clinical significance. The expression of ADAMTS-12 increased gradually with the increase of degeneration degree of human lumbar intervertebral disc.
