EPO治疗对大鼠脊髓损伤后脊髓运动功能恢复的实验性研究

【摘要】:目的:探讨人重组促红细胞生成素(rh-EPO)在大鼠脊髓损伤(SCI)后运动功能恢复中的作用。方法:选取30只200~230g成年雄性SD大鼠,随机数字表法分为三组:假手术(Sham)组、脊髓损伤(SCI)组和EPO组,每组10只。Sham组大鼠仅行椎板切除术,不伤及脊髓。SCI组和EPO组大鼠行椎板切除和脊髓打击术。术后E PO组连续7天腹腔注射rh-EPO,SCI组和Sham组采用相同剂量的生理盐水。应用BBB评分评估大鼠术后第1、3、7、14天的后肢运动功能恢复情况。HE染色观察术后第14天脊髓形态学变化、免疫荧光测定新生神经元的增殖情况。结果:SCI术后第1、3天SCI组和EPO组之间BBB评分差异无统计学意义;术后第7、14天EPO组BBB评分明显高于SCI组(P0.05,P0.01)。SCI组可见大量胶质瘢痕和空洞形成;EPO组上述改变较SCI组减轻。相较于Sham组,SCI组和EPO组新增殖神经元数目逐渐增多,各组间比较差异有统计学意义(P0.01)。结论:大剂量rh-EPO的治疗可减轻脊髓内的空洞面积、刺激脊髓新生神经元的增殖和促进大鼠脊髓损伤后后肢运动功能的恢复。
[Abstract]:Aim: to investigate the role of recombinant human erythropoietin (rh-EPO) in the recovery of motor function after spinal cord injury (SCI) in rats. Methods: 30 adult male SD rats were randomly divided into three groups: sham operation group (Sham), spinal cord injury (SCI) group and EPO group (10 rats in each group). The rats in Sham group were treated with laminectomy only. No spinal cord injury. Laminectomy and spinal cord attack were performed in SCI and EPO groups. The E PO group was injected intraperitoneally with the same dose of normal saline as Sham group for 7 days after operation. BBB score was used to evaluate the recovery of motor function in the hind limbs of rats on the 1st and 7th day after operation. The morphological changes of spinal cord were observed by HE staining on the 14th day after operation. The proliferation of neonate neurons was measured by immunofluorescence. Results: there was no significant difference in BBB score between SCI group and EPO group on the 1st day after SCI, and the BBB score in EPO group was significantly higher than that in SCI group on 714 days after SCI (P0.05 + P0.01). SCI group). The above changes in EPO group were less than those in SCI group. Compared with Sham group, the number of new proliferative neurons in SCI group and EPO group increased gradually, and the difference was statistically significant (P0.01). Conclusion: the treatment of high dose rh-EPO can reduce the area of syringomyelia, stimulate the proliferation of new neurons in spinal cord and promote the recovery of motor function of hind limbs after spinal cord injury in rats.
