基质金属蛋白酶-28在人退变腰椎间盘组织中的表达及意义

【摘要】:目的:通过实验研究探讨人退变腰椎间盘髓核组织中MMP-28的表达及变化情况,从而评价在人腰椎间盘退变疾病过程中MMP-28所扮演的角色及作用,并分析退变等级与其关联性。方法:所有患者术前均行腰椎MRI等检测,根据影像学资料及术中所见将腰椎间盘退变分为:正常组、突出组、脱出组及游离组。实验通过苏木精-伊红染色法(hematoxylin-eosin staining HE)去观察正常椎间盘组织及退变椎间盘组织的形态差异,通过免疫组织化学技术(Immunohis to chemistry IHC)检测观察MMP-28处于腰椎间盘髓核组织中的部位,使用Western-blot检测MMP-28在各个等级退变椎间盘中的蛋白表达水平。全部标本均按照选取标准取材,且所取组织标本均来源于腰椎间盘纤维环内的髓核,尽可能的避免及排除椎管内其余物质对髓核标本组织的影响,使实验更能够客观真实地反映髓核中MMP-28的表达及变化状况,实验结果数据更加可靠且具有说服力。P0.05具有统计学上显著差异性。结果:苏木精-伊红染色法结果显示正常组及退变组的细胞数目及形态均有明显差异。免疫组化结果显示MMP-28在腰椎间盘髓核细胞中的定位为胞浆内,在正常组及退变组中均有表达,但退变组细胞染色数量及程度均较正常组明显。Western-blot结果显示MMP-28表达于各等级椎间盘组织中,且与椎间盘退变等级存在一定相关性,各等级之间均具有差异且有统计学意义(P0.05)。结论:人类腰椎间盘髓核组织的退变过程中有MMP-28的参与,且与其表达存在关联性,同时MMP-28随着人类退变腰椎间盘组织退变等级的加重具有相同的增高趋势。
[Abstract]:Objective: to investigate the expression and changes of MMP-28 in the nucleus pulposus of human degenerative lumbar intervertebral disc, and to evaluate the role and role of MMP-28 in the process of human lumbar disc degeneration. The relationship between the grade of degeneration and its relationship is also analyzed. Methods: all patients were examined for lumbar vertebra MRI before operation. According to imaging data and intraoperative findings, lumbar intervertebral disc degeneration was divided into normal group, prolapse group and free group. The morphological differences between normal intervertebral disc tissue and degenerative intervertebral disc tissue were observed by hematoxylin-eosin staining (hematoxylin-eosin staining HE). Immunohistochemical technique (Immunohis to chemistry IHC) was used to detect the location of MMP-28 in the nucleus pulposus of lumbar intervertebral disc, and Western-blot was used to detect the protein expression level of MMP-28 in various grades of degenerative intervertebral disc. All the specimens were taken according to the selection criteria, and the tissue samples were derived from the nucleus pulposus in the lumbar intervertebral disc annulus, so as to avoid and exclude the influence of other substances in the spinal canal on the tissue of the nucleus pulposus specimen as far as possible. The experiment can reflect the expression and change of MMP-28 in nucleus pulposus objectively and truthfully. The experimental data are more reliable and persuasive. P0.05 has statistically significant difference. Results: the results of hematoxylin-eosin staining showed that there were significant differences in cell number and morphology between normal group and degenerative group. Immunohistochemical results showed that MMP-28 was localized in cytoplasm in nucleus pulposus cells of lumbar intervertebral disc, and was expressed in normal and degenerative groups. The results of Western-blot showed that MMP-28 was expressed in all grades of intervertebral disc tissue, and there was a certain correlation between the expression of MMP-28 and the grade of degeneration of intervertebral disc, but the number and degree of staining of cells in the degeneration group were more obvious than those in the normal group. There were significant differences among different grades (P0.05). Conclusion: MMP-28 is involved in the degeneration of human lumbar disc nucleus pulposus, and it is associated with its expression. Meanwhile, MMP-28 has the same increasing trend with the aggravation of degeneration grade of human lumbar intervertebral disc tissue.
