前折叠素样蛋白通过负性调节枯否细胞功能影响肝脏缺血再灌注损伤及机制研究

研究背景和目的:肝移植术和肝切除术是治疗良性和早期恶性肝脏疾病的有效手段,但是,肝移植术和行肝门阻断的肝切除术在术后将不可避免的诱发肝脏缺血再灌注损伤,影响患者术后肝脏功能的恢复,甚至会诱发患者围手术期肝功能异常或急性衰竭。因此,研究肝脏缺血再灌注损伤的发生机制及寻找潜在的干预靶点具有实际临床意义。前折叠素样蛋白(Unconventional prefoldin RPB5 interactor,URI),也被称为RNA聚合酶II第五亚基调节蛋白(RNA polymerase II subunit5-mediating protein,RMP)已被证实在糖代谢、调节基因转录,细胞凋亡,促进肿瘤转移等过程中发挥调节功能。近来又有研究发现,URI可以影响核转录因子-κB(Nuclear factor-kappa B,NF-κB)信号通路的激活从而调节Il-6的转录促进肝癌的转移。NF-κB信号通路主要由单核巨噬细胞介导,并是固有免疫反应激活过程中的重要信号通路。基于此,我们前期研究了URI在巨噬细胞系中对于NF-κB和丝裂原活化蛋白激酶(Mitogen-activated protein k...

髓敲URI鼠的构建及敲除效果鉴定
而且效果稳定(结果来自 6 对同窝配对成年小鼠)。Fig 2的肝脏 Kupffer 细胞 URI 敲除水平相对于野生型小鼠在 1/2 左右,且异。为了验证此模型 URI 的敲除仅限于髓系来源细胞,我们获取 WT脏组织,并经过 RT-PCR 后,比较 WT 组与 KO 组,肝组织 RMP 表并无明显差异,如 Fig 2f 所示。
坏死 3 个参考项,依据损伤情况,每一项评分从 0-3 分,最后 3 项)为肝损伤的 Suzuki 评分,经定量分析及统计学分析后,两组得分用现KO组得分为5.50±1.05,WT组得分为3.83±0.75,有统计学差异,P=0量评分比较如 Fig 3b 所示。Sham 组小鼠得分均为 0 分。Fig 3c 为构建管夹位置,可见夹闭后肝组织颜色发白,取下的肝标本边缘可见坏死
