低氧预处理和七氟醚预处理对成年大鼠短暂性全脑缺血的保护作用及对Jun-B mRNA表达的影响

目的:观察低氧预处理和七氟醚预处理对成年大鼠短暂性全脑缺血(Transient global cerebral ischemia,tGCI)的保护作用及对Jun-B mRNA表达的影响。方法:成年健康雄性(Sprague Dawley,SD)大鼠,48只,体重240~250g,随机分4组:全脑缺血再灌注组(tGCI组),低氧预处理(Hypoxic preconditioning,HPC)组,七氟醚预处理(Sevoflurane,SEV)组,低氧+七氟醚预处理组(H+S组),每组大鼠12只。tGCI组:用Pulsinelli四血管闭塞法(4VO)建立10 min tGCI大鼠模型;HPC组:反复暴露于减压舱内模拟海拔4000m,每天1 h,连续5 d;SEV组:大鼠接受2.4%七氟醚与100%氧气混合气体的预处理,1h/d,连续5d;H+S组:低氧预处理联合七氟醚预处理,连续5天,1 h/d。缺血再灌注4 h(T1)、24 h(T2)、7 d(T3)分别处死大鼠,制作海马组织冰冻切片,行(Fluoro-Jade B,FJB)荧光染色,观察海马组织神经细胞凋亡,通过Trizol提取海马组织...
