TGF-β1/miR-411/SPRY4和MAPK信号通路调控横纹肌肉瘤增殖与分化的机制研究

  目的:探讨TGF-β1抑制的microRNAs及其靶基因蛋白通过MAPK信号通路调节人类横纹肌肉瘤(RMS)肌分化及其分子机制,为RMS分化治疗提供新的治疗靶点。方法:(1)采用Real-time PCR技术检测TGF-β1抑制的microRNAs在不同RMS细胞株和组织样本中的表达,筛选出差异最明显、最广泛的microRNA作为研究对象。(2)利用MTT、3H胸腺嘧啶核苷掺入法检测microRNA对RMS细胞株增殖的影响。(3)构建裸鼠成瘤性模型,观察microRNA对活体肿瘤生长的影响;采用免疫组织化学染色检测瘤体ki-67蛋白表达情况。(4)运用生物信息学技术及网络资源预测miR-411-5p的靶基因(5)运用Real-time PCR技术、双荧光素酶报告系统及蛋白印迹法验证靶基因。(6)构建Elk-1-Luc、c-Jun-Luc荧光报告质粒,SPRY4-siRNA或siRNA-ctrl转染PKC?表达质粒处理的RMS细胞株,检测Elk-1-Luc、c-Jun-Luc荧光信号;PKC?表达质粒和SPRY4-siRNA/siRNA-ctrl共转染RMS细胞株,检测P-p38蛋白的变...
