MMP-2和TIMP-2在激光诱导形觉剥夺的高度近视豚鼠脉络膜新生血管中的表达变化
[Abstract]:Objective to study the role of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor-2 of metalloproteinase (TIMP-2) in the formation of high myopia (CNV) in guinea pigs with high myopia by establishing a choroidal neovascularization model. Methods 72 two-week-old tricolor guinea pigs were randomly divided into two groups: the experimental group (n = 36) and the control group (n = 36). The right eye of the experimental group was induced by 6 weeks of form deprivation to induce high myopia. 30 guinea pigs in each group were treated with 532 nm laser photocoagulation to induce CNV. Immunohistochemical observation of MMP-2 and TIMP-2 expression in the photocoagulation region before and after photocoagulation: detection of MMP-2 and TIMP-2mRNA expression in (retinal pigment epithelium of retinal pigment epithelium layer by real-time PCR and MMP-2 and TIMP-2mRNA expression in chorio-scleral complexes positive for MMP-2 and TIMP-2 The integrated optical density (integral optical) and the relative expression level of mRNA were analyzed statistically. Results after laser photocoagulation, the expression of MMP-2 and TIMP-2 in the experimental group and the control group were significantly up-regulated, and the expression of TIMP-2 was significantly increased on the 21st day after photocoagulation, and the peak expression of TIMP-2 was observed on the 28th day after photocoagulation. The positive expression of MMP-2 and the relative expression of mRNA in the experimental group were significantly higher than those in the control group before and after photocoagulation. The difference was statistically significant (P0.05), while the expression of TIMP-2 in the experimental group was significantly lower than that in the control group (P0.05). There was no significant difference in the expression of TIMP-2 between the two groups at each time point after photocoagulation. Conclusion MMP-2 and TIMP-2 are closely related to the formation of CNV in high myopia. The imbalance of MMP-2 and TIMP-2 expression may be involved in the occurrence and development of CNV in high myopia.
