T2DM合并冠心病患者与单纯T2DM患者血浆差异表达微小RNA筛选

【摘要】:目的筛选2型糖尿病(T2DM)合并冠状动脉粥样硬化性心脏病(简称冠心病)患者与单纯T2DM患者血浆差异表达的微小RNA(miRNA),分析可能用于判断T2DM合并冠心病的潜在生物标记物。方法选取T2DM患者60例,根据是否合并冠心病分为T2DM合并冠心病组和单纯T2DM组,每组30例。收集两组临床资料,采用微阵列miRNA芯片筛查两组差异表达的miRNA,采用实时荧光定量RT-PCR法检测两组差异表达miRNA的相对表达量(验证微阵列芯片结果),分析差异表达miRNA与T2DM患者临床参数的关系。结果微阵列miRNA芯片数据筛选出两组存在7个差异表达的候选miRNA,即miR-765、miR-320c、miR-33b-3p、miR-21-5P、miR-22-3p、miR-20b-5p及miR-92a-3p。与单纯T2DM组比较,T2DM合并冠心病组miR-765、miR-320c相对表达量降低(P均0.01)。T2DM患者血浆miR-765、miR-320c相对表达量与Hb A1C均呈负相关(β分别为-0.379、-0.192,P均0.01),与LDL-C均呈负相关(β分别为-0.224、-0.232,P均0.05)。结论 miR-765和miR-320c在T2DM合并冠心病患者与单纯T2DM患者血浆中存在差异表达,二者表达量降低提示T2DM患者有合并冠心病的可能。
[Abstract]:Objective to screen small RNAs miRNAs differentially expressed in plasma between patients with coronary atherosclerotic heart disease (CHD) and simple T2DM patients with type 2 diabetes mellitus (T2DM), and to analyze the potential biomarkers of T2DM with coronary heart disease (CHD). Methods 60 patients with T2DM were selected, According to whether coronary heart disease is complicated or not, it is divided into two groups: T2DM with coronary heart disease group and simple T2DM group with 30 cases in each group. The clinical data of the two groups are collected. Two groups of differentially expressed miRNAs were screened by microarray miRNA chip, and the relative expression of differentially expressed miRNA was detected by real-time fluorescence quantitative RT-PCR method. The clinical parameters of differentially expressed miRNA and T2DM patients were analyzed. Results the miRNAs with seven differentially expressed miRNAs were screened by microarray miRNA chip data, namely, miR-765miR-320canmiR-33b-33b-3pnmiR-21-5PnmiR-22-3pnmiR-20b-5p and miR-92a-3p.Compared with T2DM group, the relative expression of miR-765miR-320c was decreased in T2DM patients with coronary heart disease (P < 0.01). The relative expression of miR-765miR-320c and miR-765miR-320c in T2DM patients with coronary heart disease were significantly lower than those in T2DM group (P < 0.05). HbA1C was negatively correlated (尾 = -0.379- 0.192P, P = 0.01g, P = -0.224- 0.232P, P = 0.050.Conclusion there is a difference in the expression of miR-765 and miR-320c in plasma between T2DM patients with coronary heart disease and those with T2DM alone. The decrease of the expression of both suggests that type 2 DM patients may be associated with coronary heart disease.
