高脂联合维生素D3饮食对大鼠动脉组织AUF-1及IL-35表达的影响

【摘要】:目的:探讨高脂联合维生素D3饮食对大鼠动脉组织AU碱基富集区结合因子1(AUF1)及白细胞介素35(IL-35)表达的影响。方法:SD大鼠20只随机分入实验组和正常组,每组10只。实验组进行高脂饲料联合维生素D3喂养12周,取其动脉组织,以正常饲养大鼠动脉组织为对照,常规HE染色观察各组动脉组织病变情况,用实时定量PCR法分别检测动脉组织AUF1及IL-35的mRNA表达;蛋白质免疫印迹法分别检测动脉组织AUF1及IL-35蛋白表达。结果:实验组动脉组织出现轻微病变,血脂异常,AUF1存在mRNA表达上调(P0.01);炎症抑制因子IL-35 mRNA存在表达上调。(P0.01)。各组AUF1及IL-35蛋白未见高浓度表达。结论:高脂高钙饮食能诱导大鼠动脉炎症,但不足以短期内使大鼠形成典型动脉粥样硬化斑块。AUF1和IL-35可作为未来治疗炎症的治疗靶点。
[Abstract]:Objective: to investigate the effects of hyperlipidemia combined with vitamin D3 diet on the expression of AU-AUF1) and IL-35 in rat arterial tissue. Methods: twenty SD rats were randomly divided into experimental group and normal group. 10 rats in each group were fed with high fat diet combined with vitamin D 3 for 12 weeks. The arterial tissue was taken from the experimental group, and the arterial tissue of the normal feeding rats was used as the control. The pathological changes of the arteries in each group were observed by routine HE staining. The mRNA expression of AUF1 and IL-35 in arterial tissues was detected by real-time quantitative PCR, and the expression of AUF1 and IL-35 in arterial tissues was detected by Western blot. The expression of mRNA was up-regulated in AUF1, and the expression of IL-35 mRNA was up-regulated. There was no high expression of AUF1 and IL-35 in all groups. Conclusion: high fat and high calcium diet can induce arterial inflammation in rats. But it is not enough to form typical atherosclerotic plaques. AUF1 and IL-35 in the short term can be used as therapeutic targets for the treatment of inflammation in the future.
