FTY720对糖尿病大鼠心肌微血管病变的保护作用

【摘要】:目的研究1-磷酸鞘氨醇受体激动剂芬戈莫德(FTY720)对糖尿病大鼠早期心肌微血管病变的作用。方法将Spragure-Dawley大鼠分为正常对照组、糖尿病模型组与FTY720[0.001 g/(kg·d)]治疗组。大剂量链脲佐菌素(65 mg/kg)腹腔注射制备大鼠糖尿病模型,HE、Masson染色观察心肌结构大体形态。CD34免疫组织化学观察心肌微血管密度,TUNEL免疫荧光检测分析心脏微血管内皮细胞凋亡情况。组织转化生长因子β(TGF-β)、免疫组织化学观察心肌纤维化程度。Western blot测定血管细胞黏附分子1(VCAM-1)、肿瘤坏死因子α(TNF-α)和白细胞介素6(IL-6)表达水平。结果糖尿病模型组以多次随机血糖≥16.7 mmol/L作为糖尿病模型成功的标志,本研究中大鼠糖尿病模型构建成功率在85%以上。与正常对照组相比,糖尿病模型组大鼠心肌组织微血管壁增厚,VCAM-1、TNF-α和IL-6的表达升高(P0.05);FTY720治疗组心肌VCAM-1、TNF-α与IL-6的表达明显低于糖尿病模型组(P0.05)。糖尿病模型组心脏组织微血管密度减少,凋亡指数显著增高;而FTY720可以减少凋亡,促进内皮细胞增殖。结论 FTY720可能在糖尿病大鼠心脏微血管损伤中具有一定保护作用。
[Abstract]:Objective to study the effect of 1-sphingosine phosphate receptor agonist, Fengomod FTY720, on early myocardial microangiopathy in diabetic rats. Methods Spragure-Dawley rats were divided into normal control group. Diabetic model group and FTY720 [0.001 g/(kg d] treatment group.
