微小RNA-152介导叶酸对体外血管平滑肌细胞增殖迁移的抑制作用

【摘要】:目的探究叶酸对(folic acid,FA)血小板源生长因子(PDGF-BB)诱导的血管平滑肌细胞(VSMCs)增殖迁移的影响及其机制。方法体外用PDGF-BB诱导大鼠血管平滑肌细胞系A7r5增殖迁移,分为对照组、PDGF组、PDGF+FA组、PDGF+FA+miRNA-152 inhibitor组、PDGF+FA+miRNA-152 NC组。采用实时定量聚合酶链反应(RT-PCR)检测各组VSMCs内miR-152的表达水平。采用CCK-8试剂盒检测各组VSMCs增殖水平。采用划痕实验检测各组VSMCs迁移水平。采用蛋白印迹法检测细胞增殖核抗原(PCNA)蛋白、基质金属蛋白酶9(MMP-9)的表达水平。结果 RT-PCR结果显示PDGF干预降低VSMCs miR-152表达水平,而PDGF+FA组VSMCs miR-152表达水平显著高于PDGF组(P0.05)。PDGF干预VSMCs的增殖迁移能力显著增强相对于对照组(P0.05),而PDGF+FA组的VSMCs增殖迁移能力低于PDGF组(P0.05),PDGF+FA+miR-152 inhibitor组的VSMCs增殖迁移能力强于PDGF+FA组(P0.05)。结论 miR-152参与介导FA抑制PDGF-BB诱导的VSMCs增殖迁移作用。[营养学报,2017,39(5):484-488]
[Abstract]:Objective to investigate the effect of folic acid on proliferation and migration of vascular smooth muscle cells (VSMCs) induced by platelet derived growth factor (PDGF-BBB) and its mechanism. Methods the proliferation and migration of rat vascular smooth muscle cell line A7r5 were induced by PDGF-BB in vitro, and were divided into two groups: control group (control group), PDGF-FA miRNA-152 inhibitor group (PDGF-FA miRNA-152 inhibitor group) and PDGF-FA miRNA-152 group (NC group). Real-time quantitative polymerase chain reaction (RT-PCR) was used to detect the expression of miR-152 in VSMCs. CCK-8 kit was used to detect the proliferation of VSMCs in each group. The migration level of VSMCs in each group was measured by scratch test. The expression of matrix metalloproteinase 9 (MMP-9) and proliferating nuclear antigen (PCNA A) were detected by Western blot. Results the results of RT-PCR showed that PDGF intervention decreased the expression of VSMCs miR-152. However, the expression of VSMCs miR-152 in PDGF FA group was significantly higher than that in PDGF group (P 0.05). The proliferation and migration ability of VSMCs in PDGF FA group was significantly higher than that in control group (P 0.05), while the VSMCs proliferation and migration ability in PDGF FA group was lower than that in PDGF group (P 0.05) and that in PDGF FA group was higher than that in PDGF FA group (P 0.05). Conclusion miR-152 is involved in the inhibition of VSMCs proliferation and migration induced by PDGF-BB mediated by FA. [Journal of Nutrition: 484-488]
