异丙肾上腺素预处理对大鼠心肌缺血再灌注损伤作用研究

【摘要】:目的:探究异丙肾上腺素作用于大鼠心肌缺血再灌注损伤的具体步骤,讨论相关可能机理。方法:SD大鼠32只,雄性,予以分为四组,假手术组(Sham组)、缺血再灌注组(I/R组)、异丙肾上腺素预处理组(ISO组)和选择性β2肾上腺素能受体(β2-adrenergic recepter,β2-AR)拮抗剂ICI118551+ISO-I/R组(ICI组)(随机数字法),每组8只。继大鼠左冠状动脉前降支(LAD)血流中断30min之后,予血液再灌流2h以构建成心肌缺血再灌注损伤的大鼠实验模型。苏木素-伊红染色法观测大鼠心肌组织形态学,蛋白印记杂交技术用于检出磷酸化丝/苏氨酸激酶(P-Akt)的表达高低,原位末端凋亡法及免疫组织化学方法分别对心肌凋亡率及凋亡相关成分Caspase-3的表达进行检出,血清中代表氧化应激高低的指标超氧化物歧化酶(SOD)活性、丙二醛(MDA)浓度分别经黄嘌呤氧化酶法、硫代巴比妥酸法检出。结果:ISO组与I/R组比较发现,前者大鼠心肌损害减轻明显,P-Akt的表达显著增多、心肌纤维凋亡率大大减低、Caspase-3的表达减弱突出、血清SOD活性增进、血清MDA浓度减低且均具显著性(P0.05);而ICI组与ISO组相比则提示,前者大鼠心肌损害较严重,心肌纤维P-Akt蛋白成分减少明显、细胞凋亡率大幅度增高、Caspase-3表达明显增强、血清SOD活性减弱、血清MDA浓度增高且均具有显著性(P0.05)。结论:异丙肾上腺素可对大鼠心肌缺血再灌注损伤发挥有利的保护功能,具体机理可能与刺激β2-AR及PI3K-Akt信号转导通道密切相关。
[Abstract]:Aim: to investigate the specific steps of isoproterenol on myocardial ischemia reperfusion injury in rats and discuss the possible mechanism. Methods Thirty-two male SD rats were divided into four groups. Sham group, I / R group, isoproterenol preconditioning group and ICI118551 ISO-I/R group (8 rats in each group). After the left anterior descending coronary artery (lad) blood flow was interrupted by 30min, the rat model of myocardial ischemia-reperfusion injury was established by blood reperfusion for 2 hours. The expression of phosphorylated silk / threonine kinase P-Akt was detected by hematoxylin-eosin staining and protein blot hybridization. Myocardial apoptosis rate and expression of apoptosis-related component Caspase-3 were detected by in situ terminal apoptosis method and immunohistochemical method respectively. The activity of superoxide dismutase (SOD), an indicator of oxidative stress, was detected in serum. Malondialdehyde (MDA) concentration was detected by xanthine oxidase method and thiobarbituric acid method. Results compared with I / R group, the expression of P-Akt was significantly increased, the expression of Caspase-3 was significantly decreased and the activity of serum SOD was increased. Compared with the ISO group, the serum MDA concentration in the ICI group was significantly lower than that in the ISO group, which indicated that the former group had more serious myocardial damage, the myocardial fiber P-Akt protein component was significantly decreased, and the apoptotic rate was significantly increased and the expression of Caspase-3 was significantly increased in the ICI group compared with the ISO group. The activity of serum SOD was decreased and the concentration of serum MDA was increased. Conclusion: isoproterenol may play a protective role in myocardial ischemia-reperfusion injury in rats, and the mechanism may be closely related to the stimulation of 尾 2-AR and PI3K-Akt signal transduction channels.
