大豆提取物对家兔动脉粥样硬化斑块中MMP-9及TIMP-1表达的影响

【摘要】：目的:探讨大豆提取物(SE)对家兔动脉粥样硬化(As)斑块中基质金属蛋白酶-9(MMP-9)及金属蛋白酶组织抑制剂-1(TIMP-l)表达的影响。方法:取本课题组前期动物实验中制备的家兔颈动脉粥样硬化血管组织蜡块40件,其中,实验对照组、As模型组及SE低、中、高剂量组每组各8件,采用免疫组化法检测粥样硬化斑块中MMP-9和TIMP-l的表达。结果:与实验对照组相比,As模型组MMP-9和TIMP-1表达量明显升高,TIMP-1/MMP-9比值明显降低(p0.05);而与As模型组相比,SE低、中、高剂量组MMP-9和TIMP-1表达量不同程度降低,TIMP-1/MMP-9比值不同程度升高(p0.05)。经Pearson相关性分析,SE剂量与家兔颈动脉粥样硬化斑块中MMP-9和TIMP-1表达量之间呈负相关,相关系数(r)分别等于-0.720、-0.831(p0.05),与TIMP-1/MMP-9比值之间呈正相关,相关系数(r)等于0.870(p0.05)。结论:SE可促使高同型半胱氨酸血症(HHcy)诱导的As家兔颈动脉粥样硬化斑块中MMP-9和TIMP-1的表达下调,引起TIMP-1/MMP-9比值升高;SE剂量与HHcy诱导的动脉粥样硬化家兔颈动脉粥样硬化斑块中MMP-9和TIMP-1的表达量以及TIMP-1/MMP-9比值之间存在一定的剂量效应关系。
[Abstract]:Aim: to investigate the effects of soybean extract (SEE) on the expression of matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of metalloproteinase-1 (TIMP-l) in atherosclerotic plaque of rabbits. Methods: forty rabbit carotid atherosclerotic tissue wax blocks were prepared in the previous animal experiment of our group. Among them, the experimental control group had 8 wax lumps in each group with low as model and low SE, middle and high dose groups, and the control group was divided into two groups: the control group, the middle dose group, and the high dose group. The expression of MMP-9 and TIMP-1 in atherosclerotic plaques was detected by immunohistochemical method. Results: compared with the experimental control group, the expression of MMP-9 and TIMP-1 in the as model group was significantly increased and the ratio of TIMP-1 / MMP-9 was significantly decreased compared with that in the as model group, while the expression of MMP-9 and TIMP-1 in the high dose group was lower than that in the as model group, and the ratio of MMP-9 and TIMP-1 / MMP-9 was increased to different degrees in the high dose group. By Pearson correlation analysis, there was a negative correlation between the dose of SE and the expression of MMP-9 and TIMP-1 in carotid atherosclerotic plaques in rabbits. The correlation coefficient was r = -0.720 ~ (-1) -0.831% p0.05, respectively, and positively correlated with the ratio of TIMP-1 / MMP-9, the correlation coefficient (r) was 0.870 ~ (-1) p _ (0.05). Conclusion the expression of MMP-9 and TIMP-1 in atherosclerotic plaques of as rabbits induced by hyperhomocysteinemia can be down-regulated by% SE, and the expression of MMP-9 and TIMP-1 in atherosclerotic plaques of as rabbits can be down-regulated. There was a dose-effect relationship between the increase of TIMP-1 / MMP-9 ratio and the expression of MMP-9 and TIMP-1 in atherosclerotic plaque induced by HHcy and the ratio of TIMP-1 / MMP-9.
