髓样分化因子88基因rs7744多态性与冠状动脉粥样硬化性心脏病易感性及严重程度的关系

【摘要】:目的探讨中国北方汉族人群髓样分化因子88(MyD88)基因3’非编码区(3’-UTR)rs7744多态性与冠状动脉粥样硬化性心脏病(CAD)发病风险及严重程度的关系。方法收集2013年9月至2015年9月中国医科大学附属第一医院血液标本810例。CAD组540例,对照组270例,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测rs7744多态性,并收集临床资料。结果 MyD88基因rs7744符合Hardy-Weiberg平衡(P0.1)。对MyD88基因rs7744多态性与CAD发病风险的关系进行总体和分层分析。MyD88基因rs7744多态位点,年龄50岁,AG突变杂合型携带者CAD的发病风险是AA野生纯合型携带者的0.38倍(95%CI:0.17~0.93;P=0.029)。对My D88基因rs7744多态性与CAD严重程度的关系进行冠状动脉病变支数及改良Gensini评分分析。在改良Gensini评分分析中,My D88基因rs7744多态位点,GG突变纯合型携带者的改良Gensini评分显著低于AA野生纯合型携带者(5.23±3.85 vs 7.49±4.96;P=0.011)。结论 MyD88基因3’-UTR区rs7744多态性与CAD发病风险及严重程度存在关联关系。携带rs7744突变基因型的年龄50岁人群,CAD的发病率比较低。rs7744突变基因型携带者的改良Gensini评分显著低于野生型携带者。
[Abstract]:Objective to investigate the relationship between the 3'noncoding region of myeloid differentiation factor 88 (MyD88) gene and the risk and severity of coronary atherosclerotic heart disease (CAD). Methods from September 2013 to September 2015, rs7744 polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique in 810 blood samples from the first affiliated Hospital of China Medical University, 540 cases in the CAD group and 270 cases in the control group. Clinical data were collected. Results MyD88 rs7744 was consistent with Hardy-Weiberg equilibrium P0.1. The relationship between the rs7744 polymorphism of MyD88 gene and the risk of rs7744 was analyzed. The polymorphic loci of MyD88 gene. The age of 50 years old carriers with AG mutation was 0.38 times as high as 95% 95% of wild homozygous carriers. The relationship between rs7744 polymorphism of my D88 gene and severity of CAD was analyzed by coronary artery disease branch number and modified Gensini score. In the modified Gensini score analysis, the modified Gensini score of homozygous carriers with rs7744 polymorphism of my D88 gene was significantly lower than that of carriers with AA wild homozygotes (5.23 卤3.85 vs 7.49 卤4.96). Conclusion there is an association between the rs7744 polymorphism in the 3- UTR region of MyD88 gene and the risk and severity of the disease. The incidence of rs7744 in 50 years old people with rs7744 mutation genotype was lower than that in wild type carriers.
