牛蒡根水提物对高血压血清致伤血管内皮细胞TLR4、NF-κB表达的影响

【摘要】:目的:探究牛蒡根水提物对高血压患者血清致伤血管内皮细胞Toll样受体-核转录因子(TLR-NF-κB)信号转导通路的影响。方法:选取2014年12月至2016年1月于医院诊治的高血压病患者40例纳入高血压组,选择健康体检者40例纳入对照组。分别采用两组研究对象的血清干预人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC-C)24h,应用不同浓度的牛蒡根水提取物干预高血压患者血清诱导的HUVEC-C,Toll样受体4(toll like receptor 4,TLR4)表达(24h)及核转录因子(nuclear factorκB,NF-κB)活化(2h)。将细胞连续培养24h后将细胞蛋白提取,并用免疫印迹(Western blot)技术检测细胞内TLR4蛋白及NF-κB抑制因子I-κBα蛋白的表达。结果:(1)高血压组细胞提取蛋白中的TLR-4水平为(0.6683±0.07468),明显高于对照组的(0.2220±0.02749),I-κBα水平为(0.2687±0.02519),明显低于对照组的(0.5616±0.07762),差异均有统计学意义(P0.05);(2)牛蒡根水提物干预低剂量组、中剂量组及高剂量组细胞蛋白中的TLR-4表达水平为(0.2720±0.05178)、(0.4509±0.03399)、(0.3810±0.07796),均低于高血压组的(0.6683±0.07468),同时高于对照组的(0.2220±0.02749);I-κBα表达水平为(0.5059±0.03042)、(0.3433±0.02289)、(0.3653±0.00710),高于高血压组的(0.2687±0.02519),同时低于对照组的(0.5616±0.07762),差异均有统计学意义(P0.05)。结论:经TLR-NF-κB信号途径介导的免疫紊乱和炎症反应可能是高血压病血管内皮损伤的机制之一,牛蒡根水提取物可通过抑制细胞TLR-4、NF-κB的表达,来抑制炎症因子的表达,最后达到保护血管内皮细胞的作用。
[Abstract]:Aim: to investigate the effect of burdock root water extract on Toll-like receptor-nuclear transcription factor (TLR-NF- 魏 B) signal transduction pathway in serum injured vascular endothelial cells in patients with hypertension. Methods: 40 hypertensive patients treated in hospital from December 2014 to January 2016 were selected into the hypertension group and 40 healthy persons were selected as the control group. Human umbilical vein endothelial cells (human umbilical vein endothelial cells HUVEC-C) were treated with serum from two groups for 24 h. Different concentrations of Arctium lappa root water extract were used to interfere with the expression (24 h) and the activation of nuclear transcription factor (nuclear factor 魏 B) NF- 魏 B (2 h) of HUVEC-CT-Toll-like receptor 4 (toll like receptor 4 (TLR4) and the activation of nuclear transcription factor (nuclear factor 魏 B (NF- 魏 B) in hypertensive patients. After 24 hours of continuous culture, the cell protein was extracted, and the expression of TLR4 protein and NF- 魏 B inhibitor I- 魏 B 伪 protein was detected by Western blot. Results: (1) the level of TLR-4 in cell extract protein in hypertension group was (0.6683 卤0.07468), significantly higher than that in control group (0.2220 卤0.02749) and (0.2687 卤0.02519), significantly lower than that in control group (0.5616 卤0.07762).
