miR-4463在下肢动脉硬化闭塞症中的表达及意义

【摘要】:目的观察microRNA-4463(miR-4463)在下肢动脉硬化闭塞症(ASO)患者血浆和组织中的表达变化,推测其可能意义。方法采用miRNA芯片方法筛选ASO患者和对照人群血浆中差异表达miRNA,Real time PCR技术验证ASO患者和对照人群中miR-4463的表达水平,并进行临床分期比较。靶基因预测软件分析miR-4463靶基因,Gene Ontology和KEGG数据库分析靶基因的功能和信号通路。结果与对照组相比,ASO患者血浆中有51个miRNA变化超过1.5倍(P0.05)。miR-4463在ASO患者血浆和病变血管内膜组织中表达均显著降低,并随Fontaine分期呈逐渐下降趋势。生物信息学分析发现miR-4463靶基因与细胞迁移、脂质代谢、内吞作用等相关。结论 miR-4463参与了ASO的病程,其表达下降可能提示ASO的发生。
[Abstract]:Objective To observe the changes in the expression of microRNA-4463 (miR-4463) in the plasma and tissue of patients with arteriosclerotic occlusive disease (ASO) of the lower extremities, and to speculate on its possible significance. Methods the differential expression of miRNA in the plasma of ASO patients and the control population was screened by miRNA chip method. The Real time PCR technique was used to verify the expression level of miR-4463 in the ASO patients and the control population. The target gene prediction software analyzed the miR-4463 target gene, the Gene Ontology and the KEGG database to analyze the function and signal pathway of the target gene. Results compared with the control group, 51 miRNA changes in the plasma of ASO patients were more than 1.5 times (P0.05).MiR-4463 in the plasma of ASO patients and the intima tissue of the lesion vessels significantly. Fontaine staging was gradually decreasing. Bioinformatics analysis found that the miR-4463 target gene was related to cell migration, lipid metabolism, endocytosis, etc. conclusion miR-4463 participates in the course of ASO, and the decline of its expression may suggest the occurrence of ASO.
