血管内皮生长因子165对心肌样细胞活性的影响

【摘要】:目的:探讨携带血管内皮生长因子165(vascular endothelial growth factor 165,VEGF165)基因的腺病毒(Ad-VEGF165)转染至心肌样细胞对其表达VEGF165及增殖活性的影响。方法:提取、培养、鉴定骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs);体外诱导BMSCs为心肌样细胞。构建携带VEGF165基因的腺病毒载体并转染至心肌样细胞;实验分为3组:Ad-VEGF165转染心肌样细胞作为实验组;未携带VEGF165基因的腺病毒转染心肌样细胞作为空载体组;未处理的心肌样细胞作为空白对照组。在1周内检测各组VEGF165的表达及心肌样细胞的增殖活性和相关指标表达。结果:VEGF165在实验组可正常表达,与空载体组、对照组相比差异具有统计学意义(P0.05);心肌样细胞增殖活性在实验组中最佳,显著优于空载体组和对照组(P0.05)。结论:成功将Ad-VEGF165转染至心肌样细胞,且表达的VEGF165可增强心肌样细胞的活性。
[Abstract]:Aim: to investigate the effect of adenovirus (Ad-VEGF165) carrying vascular endothelial growth factor 165 (VEGF 165) gene on the expression and proliferation of VEGF165 in cardiomyoid cells. Methods: bone marrow mesenchymal stem cells (bone marrow mesenchymal stem cells) were isolated, cultured and identified. The adenovirus vector carrying VEGF165 gene was constructed and transfected into cardiomyocyte-like cells. Untreated cardiomyoid cells served as blank control group. The expression of VEGF165, the proliferative activity of cardiomyocyte-like cells and the expression of related indexes were detected within 1 week. Results the expression of VEGF165 was normal in the experimental group, and the difference was statistically significant compared with the empty vector group and the control group (P0.05), and the proliferation activity of myocardial like cells was the best in the experimental group, which was significantly better than that in the empty vector group and the control group (P0.05). Conclusion: Ad-VEGF165 was successfully transfected into cardiomyocyte-like cells, and the expression of VEGF165 could enhance the activity of cardiomyocyte-like cells.
