轻型地中海贫血成年男性精液质量及其精子γ-H2AX含量的研究

【摘要】:目的研究轻型地中海贫血成年男性精液质量,探讨精子γ-H2AX含量及其与精液参数、血清铁蛋白浓度的关系。方法选取轻型地中海贫血的成年男性100例(轻型地中海贫血组)和确认有生育能力的健康男性100例(对照组)。通过检测精子γ-H2AX含量、精子DNA损伤率、血清铁蛋白浓度、精浆活性氧物质(reactive oxygen species,ROS)及精液常规参数,分析两组间差异及轻型地中海贫血组精子γ-H2AX与精液参数、血清铁蛋白浓度的相关性。结果相比对照组,轻型地中海贫血组的精子γ-H2AX表达率、精浆ROS、DNA损伤率及血清铁蛋白浓度显著增高(P0.001),而精子浓度、精子前向活动力及精子正常形态百分率显著降低(P0.001);精子γ-H2AX表达率与精子DNA损伤率、精浆ROS水平、血清铁蛋白浓度呈正相关(P0.001),而与精子前向活动力、精子正常形态百分率呈负相关(P0.001)。结论轻型地中海贫血成年男性的精液质量较健康男性差,γ-H2AX含量明显高于健康男性,这可能与铁负荷增加所导致的精子DNA损伤率升高有关。
[Abstract]:Objective to study the semen quality of adult male patients with mild thalassemia and to explore the relationship between sperm 纬-H2AX content and semen parameters and serum ferritin concentration. Methods 100 adult males with mild thalassemia (mild thalassemia group) and 100 healthy men (control group) were selected. The sperm 纬-H2AX content, sperm DNA damage rate, serum ferritin concentration, seminal plasma reactive oxygen species (reactive oxygen species,ROS) and semen routine parameters were measured to analyze the difference between the two groups and the sperm 纬-H2AX and semen parameters in mild thalassemia group. Correlation of serum ferritin concentration. Results compared with the control group, the sperm 纬-H2AX expression rate, seminal plasma ROS,DNA damage rate and serum ferritin concentration in the mild thalassemia group were significantly higher than those in the control group (P0. 001). The percentage of forward motility and normal morphology of spermatozoa decreased significantly (P0.001). The expression rate of 纬-H2AX was positively correlated with sperm DNA damage rate, seminal plasma ROS level and serum ferritin concentration (P0. 001), but negatively correlated with sperm forward motility and the percentage of normal sperm morphology (P0. 001). Conclusion the semen quality of adult males with mild thalassemia is worse than that of healthy men, and the content of 纬-H2AX is significantly higher than that of healthy men, which may be related to the increase of DNA damage rate of sperm caused by the increase of iron load.
