冠心病患者外周血单个核细胞EPC百分比及SDF-1、VEGF-2水平变化及意义

【摘要】:目的探讨冠状动脉粥样硬化性心脏病(简称冠心病)患者外周血单个核细胞内皮祖细胞(EPC)百分比及血清基质细胞衍生因子1(SDF-1)、血管内皮生长因子2(VEGF-2)水平变化及意义。方法选取冠心病患者91例(冠心病组),体检健康者40例(对照组)。采用流式细胞术测定外周血单个核细胞EPC百分比,ELISA法检测血清SDF-1、VEGF-2;分离冠心病患者单个核细胞,分别给予0、10、20、40、80、160 ng/m L CXC族趋化因子受体4(CXCR4,SDF-1受体)拮抗剂AMD3100共培养7天,观察各浓度AMD3100对EPC百分比及SDF-1、VEGF-2水平的影响。结果冠心病组外周血单个核细胞EPC百分比及SDF-1、VEGF-2水平均低于对照组,其中Ⅱ型斑块冠心病患者三项指标均低于Ⅰ型和Ⅲ型患者,三支冠状动脉狭窄患者三项指标均低于单支和双支患者,冠状动脉侧支循环2、3级冠心病患者三项指标均高于0、1级患者,组间及组内比较P均0.05。随着AMD3100浓度的升高,单个核细胞EPC百分比及血清SDF-1、VEGF-2水平依次降低,各浓度间比较P均0.05。结论冠心病患者外周血单个核细胞EPC百分比及血清SDF-1、VEGF-2水平均降低,三者的水平变化与冠心病的发生、发展有关;通过SDF-1动员EPC、VEGF-2,修复受损血管和形成新生血管,有望成为临床治疗冠心病的新靶点。
[Abstract]:Objective to investigate the percentage of endothelial progenitor cells (EPC) in peripheral blood mononuclear cells (PBMC) and serum stromal cell derived factor-1 (SDF-1) in patients with coronary atherosclerotic heart disease (CHD). Changes and significance of vascular endothelial growth factor 2 (VEGF-2) level. Methods 91 patients with coronary heart disease (CHD group) and 40 healthy persons (control group) were selected. The percentage of EPC in peripheral blood mononuclear cells was measured by flow cytometry and serum SDF-1,VEGF-2; was detected by ELISA method. Mononuclear cells were isolated from patients with coronary heart disease and were co-cultured for 7 days with AMD3100, a chemokine receptor 4 (CXCR4,SDF-1 receptor) antagonist of CXC group. The effect of AMD3100 concentration on EPC percentage and SDF-1,VEGF-2 level was observed.
